| There are many natural plants, Chelidonium majus L. is widely distributed among active substance in China, the major active substances including chelidonine, chelerythrine and berberine base, with a variety of biological activity. In this study, to choose the Chelidonium majus L. as research subjects, and macroporous resin extraction and separation of the crude extract enriched them to 3rd instar larvae of gypsy moth as the test insect larvae, by toxicity measurement, calculated virulence regression equation, LC50 and chi-square value, and with 95% efficient cyfluthrin, imidacloprid 93.8% and 94.3% acetamiprid three chemical agents twenty-two complex, wherein the synergistic combination screened and determined the best ratio synergistic combination aimed proven synergy of botanical pesticides, reduce the use of conventional pesticides, for the development of a safe, efficient, good environmental compatibility of new botanical pesticides to provide a theoretical basis.By soxhlet extraction, cold soak and ultrasonic extraction prepared Chelidonium majus L. crude extract, and stomach by law and tag method to 3rd instar larvae of gypsy moth indoor virulence test results showed that the three methods of treatment when crude extract both stomach poisoning activity or contact activity effects are general, and larval death more slowly, usually in the 48 h after a slight effect, where the effect is slightly better soxhlet extraction, liquid concentration of 250 μg/mL, stomach poison the best, the mortality rate reached 36.67%, when the liquid concentration 200 μg/mL, the best tag, the mortality rate reached 38.33%.In order to further separation and purification and enrichment Chelidonium majus L. crude extract of chelidonine, select nine kinds of different physical and chemical properties of macroporous resin adsorption test. Static and dynamic adsorption and desorption of adsorption, elution gradient and isocratic filter out the best macroporous resin. The results show, D-101 resin significantly better than several other macroporous resin, the final adsorption conditions: the sample solution 300 mL (30BV), a flow rate of 1.0 mL, pH value 2.0, temperature 25℃; gradient elution conditions:20% ethanol,40 mL (4BV), then 70% ethanol 120 mL (12BV), a flow rate of 1.0 mL. Macroporous resin can be further isolated and purified chelidonine extraction rate increased from 2.67% to 21.48%.Based on the above tests, of chelidonine combined virulence effect, choose their own separation and purification of 21.48% chelidonine and 95% efficient cyfluthrin, imidacloprid 93.8% and 94.3% as the original drug Acetamiprid the measurement results through laboratory virulence calculated cooperative virulence index (c.f). Filter out 21.48% chelidonine+94.3% diflubenzuron synergistic combination of this group, the synergistic combination of virulence index (c.f) of 21.46, and then determine the best use of toxicity ratio index ratio synergistic combination, and found to 1:4, the synergic effect of the best, toxicity ratio reached 1.21, in order to reduce the use of traditional chemical pesticides may be provided. |
