II Some in vitro biological properties of a porcine Lactobacillus acidop Hilus were evaluated in this study,and selected the best formulation of protectant for it.Firstly sterilization soybean meal,ordinary soybean meal and lactation feeds for sows were fermented by a lyop Hilized dosage form,and then the optimal fermentation parameters of three kinds of matrix were optimized based on the previous data.Additionally,the nutritional changes were analyzed before and after fermentation.And we also carried out an experiment using fermented feed by porcine Lactobacillus acidop Hilu fed sows to verify the real efficiency.1.The results showed that the strain exhibited optimal growth situation when the inoculate dose was 4%.The content of lactic acid was the peak of 66.85 mmol/L when culture 60 hours.The survival rate of the strain was 70.14% when incubated at p H=2.0 for 4 hours.And in the condition of 0.2% and 0.3% of pig bile salts,the number of living bacterium were respectively 6.11 and 4.53 log(CFU/m L)for 2 hours.The inhibitory zone diameter restrained Escherichia coli,Salmonella and Stap Hylococcus aureus was above 19 mm.When the concentration of bacterial suspension was 109CFU/m L,the adhesion index of the strain in intestinal porcine epithelial cells reached the peak of 1060CFU/100 cell.2.The formulation of protectant for lyop Hilized Lactobacillus acidop Hilus was optimized by uniform experiment.The results indicated that the optimal protectant formula were 16.216 skim milk,4.375% lactose,0.123% L-monosodium glutamate,0.143%L-Cysteine and 9.469% dextrin.The survival rate of this strain after freeze drying could reach 93.91% using the optimal compound protectant.3.The optimal fermentation conditions for sterilization soybean meal were 37? fermentation temperature,1:1.2 ration of material to water,0.8% inoculumsize,36 h fermentation time,and 9.10 lg(CFU/g)living Lactobacillus acidop Hilus.The optimal fermentation conditions for ordinary soybean meal were 37? fermentation temperature,1:1.2 ration of material to water,1.0% inoculumsize,48 h fermentation time,and 9.02 lg(CFU/g)living Lactobacillus acidop Hilus.The optimal fermentation conditions for lactation feeds for sows were 37? fermentation temperature,1:0.8 ration of material to water,0.4% inoculumsize,36 h fermentation time,and 9.48 lg(CFU/g)living Lactobacillus acidop Hilus.After fermentation,the number of living bacterium in sterilization soybean meal,ordinary soybean meal and the compound feed were changed from 106 CFU/g to 109 CFU/g,and the volatile base nitrogen content were less than 50mg/100 g.Fortunately,the crude protein content respectively increased 2.16%,1.80% and 13.10%.The acid soluble protein content respectively increased 10.36%,11.73% and 20.62%.The calcium content respectively increased 3.13%,3.13% and 3.23%.The total p Hosp Horus content respectively increased 2.78%,2.82% and 4.92%.4.Sow feeding experiment results showed that adding solid state fermentation feed improve the rate of born and weaned,the average daily gain of piglets 2.60%,9.20% and 9.10%,respectively.And the piglets' diarrhea rate reduced 53.24%.The average daily feed intake of lactation sows rose 13.79%.In addition,the Ig G content in sows' serum promoted 7.58%,and the protein,total solids,Ig A and Ig M in colostrums raised 18.08%,1.30%,4.06% and 5.61% as well.The quantity of lactic acid bacteria in fresh feces of lactation sow was changed from 7.59 to 9.04 lg(CFU/g).
