| China is the center of the origin and distribution of Chrysanthemum morifolium.Although chrysanthemum is abundant in germplasm resources,various in cultivars and rich in flower forms and colors,but are mainly focused on its ornamental value,have less research on food value.Therefore,this study selected Chrysanthemum varieties of Nanjing Agricultural University“Chinese chrysanthemum germplasm conservation center" provides as a test material,to explore its nutritional value as well as flowers or leaves content of zinc and selenium by spraying of zinc fertilizers(ZnSO4·7H2O)and selenium fertilizer(NaSeO3),by the optimum spraying concentration conducted transcriptome selected inquiry.The main findings are as follows:1.By using different concentrations of foliar application of selenium fertilizer,zinc fertilizers,selenium zinc interaction fertilizer to study leaves of nutrients as well as zinc and selenium content after spraying fertilizer on Chrysanthemum nankingense.It was found that the best soluble sugar content significantly increased 68.91%than the control 0.02g/L concentration of selenium fertilizer spraying effect,but soluble protein content cf.decrease 17.85%compared with the polyphenol content is 60 times higher,and selenium utilization rate of 0.94%,however,mutually reinforcing effect of selenium and Zn fertilizer is indistinct.2.Spraying different concentrations of selenium fertilizer(NaSeO3),zinc fertilizers(ZnSO4·7H2O),selenium zinc interaction fertilizer on ’Lollipop purple,and ’Zembla Sunny’ nutrients and flowers of zinc and selenium content.By measuring physiological parameters associated with the content of zinc and selenium,then comparing the content of soluble sugar,soluble protein,Polyphenol,total flavone,ehiorogenie acid in order to conclude that ’Lollipop purple’overall level higher than ’Zembla Sunny’ reanalysis’Lollipop purple,respective spraying concentration,and Se3 is best,soluble sugar content is processed Sel three times,soluble protein content is about 4 times of CK,Polyphenol compared with about three times higher processing Se2,CK is more than 10 times,total flavone content is 2 times CK and chlorogenic acid content is 2 times CK,so that the spraying programs and selected varieties for the single application of selenium 0.06g/L concentration ’Lollipop purple’.3.Sequenced by Illumina HiSeq TM 2000 platform,differences in gene analysis aspect of edible chrysanthemum ’Lollipop purple’,according to a large number of transcriptome data obtained in selenium treatment and water treatment analysis were obtained two libraries,produce clean reads 159,537 with a total bases 40.38Gb,and analyzed the related enzymes expression under different treatment conditions.It was found that Selenoamino and selenium-binding protein related genes has differentially expressed in ’Lollipop purple’flower after Se treatment,treatment compared with the control up-regulated.The application of se helps protein synthesis in Chysanthemum morifolium.Also found chlorophyll reductase NCY1 gene down-regulated associated with plant stress resistance.By slowing the degradation of chlorophyll process thus enhancing the synthesis of chlorophyll,increase plant resistance. |
