| Efficient coolant,a new-type convenient and environmentally friendly physical cooling material,could be applied to the postharvest cold-chain transportation and storage of the cut flowers.So far,there is little relevant researches and literatures report on the fresh-keeping effects of the efficient coolant on the cut flowers and their mechanism.Also,no relative technologies application of the efficient coolant on the postharvest transportation and storage and fresh-keeping of cut flowers.Thus,this thesis firstly studied and established a method for simulative transport and storage with selected coolant of cut roses(Rosa hybrid ?Movie star?)and spary carnations(Dianthus caryophyllus ?Prince?).Then focused on the change in ornamental quality and vase life after simulative transport and storage and during vase period,and the characteristics of some relevant postharvest physiological indexes of cut roses ?Carola? with appropriate low-temperature of coolant treatment.Finally based on the RNA-seq analysis,the main expression laws of some genes in the petals of cut roses.The major results were as follows:1.The feasibility and basic method of simulative transport and storage for cut roses ?Movie star? and cut spary carnations ?Prince? with a selected new-type efficient coolant.The result showed that:(1)The coolant had excellent effect of cold accumulation in the this study.The treatment with appropriate low-temperature(-13.5℃ and-19.3℃)coolant could continually maintain the inner temperature(2℃)of the warm-box during the 48 h period of the simulative transport and storage.Also it is helpful to maintain a higher relative humidity(about 92%)in the warm-box;(2)It had obvious fresh-keeping effects of the cut roses ?Movie star? and cut spray carnations ?Prince? with appropriate low-temperature coolant treatment,namely could prolong the longevity,improve ornamental quality and maintain the relative fresh weight of the both cut flowers.And the best low-temperature of coolant treatment used in the cut roses ?Movie star? and cut spray carnations ?Prince? were-13.5℃ and-19.3℃,respectively.2.The fresh-keeping effects of cut roses ?Carola? were further investigated using the established method of simulative transportation and storage with coolant for cut roses ?Movie star?.The result showed that:(1)The coolant treatment could prolong significantly the longevity and improve ornamental quality of cut roses ?Carola?.(2)The relative fresh weight,water up-take,water loss and water balance value of cut roses ?Carola? treated with the coolant were all better than those of the control.In addition,the water content of petal of cut roses ?Carola? was obviously higher than the control during 48 hours in coolant treatment and succedent vase period.(3)The coolant treatment had benefit to maintain the anthocyanin content int the petals of cut roses.It had no significant difference in chlorophyll content of leaves of cut roses between the coolant treatment and control,though the chlorophyll content of the cooltant treatment was somewhat lower than that of the control immediately after 48 h low-temperature simulative transport and storage.3.There were 96270 Unigenes and 49998 annotations in the results of RNA-seq of petasl of cut roses ?Carola?.Compared with the KEGG datebase and different expresion genes(DEGs)enrichment analysis of coolant treatment and the control in the day 0 of vase period,it was found that the main pathways responding to the low temperature were ?plant hormone signal transduction?(81 DEGs),?starch and sucrose metabolism?(59 DEGs)and ?phenylpropanoid biosynthesis?(47 DEGs).The trends of Unigene expressions of the key enzymes in anthocyanin synthesis,chlorophyll synthesis and decomposition were totally in accord with the changes in the contents of petal anthocyanin and leaf chlorophyll.Additionnaly it was found that ethylene synthesis,ethylene signal transduction and cold respond C-repeat biding factor(CBF)genes were significantly up-regulated immediately after simulative transport and storage. |
