| The aim of this study was preparation of fermented Qingwenbaidu formula by two strains of Lactic acid bacteria(Lactobacillus plantarum and Enterococcus faecium)to explored effect on the growth and optimize the fermentation technology,expanded production using fermentation tank.And then studied the effects of fermentation on the performance of weaned piglets supplemented with the fermentation preparation by water solution.The experiments were as follows:(1)Effects of Qingwenbaidu formula on growth of Lactic acid bacteria and optimization of fermentation technology.Firstly,to explore its effect on the growth of Lactic acid bacteria,the experiments were divided into six groups to cultivate the two strains of Lactic acid bacteria according different volumes(0、20%、40%、60%、80%、100%)of Qingwenbaidu formula.The results showed that the population of Lactic acid bacteria was increased first and then decreased with the increase of the drug concentration and the optimum adding volumes of Qingwenbaidu formula was 40%.The result illustrated that it promote the growth of Lactic acid bacteria.Secondly,the addition content was optimized such as brown sugar,soybean meal and yeast extract in the culture medium by L9(34)three-factor and three-level orthogonal test.The results showed that yeast extract had the greatest effect on the growth of Lactic acid bacteria,followed by soybean meal and brown sugar.The optimum medium(per liter)was: brown sugar 20 g,soybean meal 5g,yeast extract 4g,CH3 COONa 3g、K2HPO4 2g、Mg SO4 0.1g、Mn SO4 0.02g、Tween 80 1m L、p H 7.0.In this case,the population of Lactic acid bacteria increased to 7.6×108 CFU/m L,which was 1.38 fold of unfermentation.Thirdly,the fermentation conditions were optimized such as initial p H,inoculation amount,fermentation temperature and fermentation time.The optimum fermentation conditions were as follows: initial p H was 7.0,inoculation amount was 2.5%,fermentation temperature was 34 ℃,fermentation time was 36 h.The population of Lactic acid bacteria increased to 1.22×109 CFU/m L,which is 2.18 fold of unfermentation.(2)Cultivation experiment and optimization of fermentation tank.The fermentation process optimized for the vials was verified by the fermentation tank test.The results showed that the amount of Lactic acid bacteria was increased by 1.32 fold.This proved that it provides a more suitable growth environment for Lactic acid bacteria and it is moreconducive to the growth of Lactic acid bacteria.Ca CO3 could promote the growth of Lactic acid bacteria,and the appropriate addition was 1.5%.Increasing the fermentation tank speed could promote the growth of Lactic acid bacteria,and suitable speed was 70 rpm.(3)The application of fermented Qingwenbaidu formula on weaned piglets.A single-factor test was used in the study and drugs were added by drinking water.360 weaned piglets(Duroc× Landrace× Large White,28(±3)days)were divided into randomly 6 groups:control group,Lactic acid bacteria oral solution group(200-fold dilution),Qingwenbaidu formula group(200-fold dilution),three fermented groups(400-fold dilution in group A,B group 200-fold dilution,group C 100-fold dilution),each group was 3 repetitions,20 repetition.The trial period was 28 days.Calculating the average feed intake,average daily gain,feed/weight ratio,diarrhea rate and elimination rate.The results showed that the fermented groups could significantly increase the average daily feed intake,average daily gain,and significantly reduce the feed weight ratio,diarrhea rate and elimination rate compared with the control group.The effect of group B and group C were significantly better than that of unfermented group and Lactic acid bacteria oral solution group.Compared with the control group,Lactic acid bacteria oral solution group and unfermented group,the average daily feed intake in group B increased by 13.2%(P<0.05),11.9%(P<0.05)and 2.5%(P<0.05)respectively,the average daily gain increased by 29.4%(P<0.05),24.9%(P <0.05)and 7.1%(P<0.05)respectively.The feed weight ratio decreased by 12.6%(P<0.05),10.4%(P<0.05)and 4.6%(P<0.05)respectively,and the diarrhea rate decreased by 72.8%(P<0.05),65.3%(P<0.05)and 40.6%(P<0.05)respectively.Compared with the control group,Lactic acid bacteria oral solution group,the elimination rate in group B decreased by 83.5%(P<0.05)and 80.2%(P<0.05)respectively.This study implied that fermented drugs increased efficacy for the medical care of weaning piglets and the optimum added dose was 200 fold dilution by water solution. |
