The low-molecular-weight glutenin subunits(LMW-GS)account for about one-third of wheat seed storage proteins and have great effects on dough extensibility and food processing quality.Common wheat and its relatives are important genetic resources for wheat improvement.In this study,the specific primers of Glu-A3 and Glu-B3 genes are used to identify and amplify the allelic variations of LMW-GS genes from twenty-one wheat varieties with different gluten contents and common wheat related species.A total of thirty-three new DNA haplotypes and twenty-six new amino-acid sequences were identified,including nineteen Glu-A3 haplotypes and nineteen amino-acid sequences on Glu-A3 loci,five Glu-B3 haplotypes and three amino-acid sequences on Glu-B3 loci,nine Glu-D3 haplotypes and four amino-acid sequences on Glu-D3 loci.Among them,the corresponding peptide of GluA3-18 from wheat variety Shiluan 02-1 and T.dicoccum DM2 contains nine cysteine residues,the corresponding peptide of GluA3-312 from wheat variety T.boeoricum B03 contains nine cysteine residues and the corresponding peptide of GluD3-13 from Yanzhan 4110 has seven cysteine residues,both of which belong to the specific subunits compared with others with eight cysteine residues,theree amino-acid sequences contained nine cysteines or seven cysteines that might be good for improvement of wheat quality.The results about Glu-3 genes in wheat and it’s related species further made clear composition of LMW-GS and provided a basis for selection of parents in wheat quality breeding.
