ObjectiveTo Lay the foundation for further exploring the function and mechanism of CLK family members in early developmental of chick embryos,the expression pattern of protein kinase CLK family(CDK-like kinase,including CLK1,CLK2,CLK3,CLK4)m RNA in early chick embryos were investigated.Methods1.The expression of CLKs m RNA were detected by q PCR.2.Molecular cloning methods was used for construction of recombinant CLKs/p MD18-T,and verified.Digoxigein labeled RNA probes of CLKswere prepared.3.To detect the distribution of CLKs m RNA in early chick embryos using the whole-mount in situ hybridization.4.To detect the location of CLKs m RNA in early chick embryos using frozen section in situ hybridization.Results1.The m RNA level of CLKs in early chick embryos changed along with the progress of different periods in chick embryos development.2.The recombinant bait plasmid CLKs/p MD18-T were constructed successfully,and preparation for digoxigin labeled RNA probe of CLKs have been done.3.The expressions of CLKs in early chick embryos are almost ubiquitous,especially in head,optic vesicles,neural folds and neural plates.ConclusionsThe m RNA level of protein kinases CLK family in early chick embryos changed along with the progress of different periods in chick embryos development.The results of in situ hybridization reveal that the expressions of CLKs are almost ubiquitous,especially in head,optic vesicles,neural folds and neural plates,suggesting that they may be associated with the development of eye and nervous system in early chick embryos. |
