Japanese encephalitis(JE)is a zoonotic infectious diseases caused by Japanese encephalitis virus.Japanese encephalitis can cause inflammation of the central nervous system,and thus induce a large number of peripheral immune cells migration to the central nervous system.These peripheral immune cells migrate into the central nervous system and release inflammatory factors to induce the secondary storm of central nervous system.However,the mechanism of peripheral immune cells migrating to the central nervous system caused by Japanese encephalitis virus is not very clear and the role of Japanese encephalitis virus in apoptosis and activation of cerebral microvascular endothelial cells is rarely reported.Based on the above backgrounds,this study investigated the effects of Japanese encephalitis virus on apoptosis and activation of brain microvascular endothelial cells and explored the physiological effects and barrier function of activated brain microvascular endothelial cells.Specific research as follows:Flow cytometry was used to detect the early apoptosis and late apoptosis of endothelial cells after JEV infection and the results showed that JEV-P3 strain virus significantly reduced the early apoptosis and late apoptosis of endothelial cells;WB and yeast expressing LFAHAGFP+was used to detect the expression of adhesion molecules on endothelial cells,and the results revealed that the expression of VE-Cadherin?ICAM-2?ICAM-1?beta-catenin were up-regulated by JEV P3 strain;The peripheral leukocyte adhere on endothelial after virus infection was detected by fluorescence microscopy,and it was found that JEV-P3 virus could increase the adhesion of endothelial cells to peripheral leukocyte;ECIS was applied to detect the barrier function of endothelial cells and the results showed that the activation of endothelial cells did not change the function;The expressions of ROS and some cytokines in endothelial cells were detected after the activation of endothelial cells.In summary,the JEV-P3 promoted the expression of adhesion molecules on brain microvascular endothelial cells,and induced the activation of brain microvascular endothelial cells and did not change its barrier function and the intercellular connection.Endothelial cells after JEV infection may increase adhesion to peripheral immune cells through IL-6 and IP-10.This study further explains the function and mechanism of the activation of cerebral microvascular endothelial by JEV,and provide new way for the treatment of JE.
