Fining Mapping And Predicting Candidate Gene Of A Major QTL ,qPH3.05, For Plant Height In Maize | | Posted on:2018-02-26 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y C Xu | Full Text:PDF | | GTID:2323330515987551 | Subject:Genetics | | Abstract/Summary: | PDF Full Text Request | | Maize(Zea mays L.)is a very important crop,mainly used as feed grain and bioenergy crop.It is of importance to increase the yield of maize.Breeders usually increase the production by improving single plant yield and planting density.A rational plant height has a profound influence on multiple,such as planting density,lodging resistance,photosynthetic efficiency and so on.Gene cloning is still an important method to analysis genetic mechanism of plant height.This study combined RNA-Seq to fine map a QTL controlling plant heigh.The main results are summarized as follows:1.We detected a major QTL on chromosome 3 controlling plant height by screening a RIL group derived from the cross between DE3 and BY815.By investigating the progenies,it was indicated this QTL is real and its confidence interval is about 1Mb.Then the target region was narrowed down to a region 600Kb;2.Based on the information of B73 Ref v3,there were 31 genes within the 1Mb region and 16 genes were at the 600 Kb region.RNA-Seq indicates that 5 genes were expressed different and 2 genes were on the 600 Kb region;3.We found 8 of 31 genes may be relevant with plant height by analyzing genes’ annotation,4 of which are located on the 600 Kb candidate regions;4.By combining gene annotation and RNA-Seq date,we predicted gene GRMZM2G177227 would be the candidate gene,and further validation is still needed. | | Keywords/Search Tags: | Maize(Zea mays L.), Plant height, RNA-Seq, Quantitative trait locus(QTL) | PDF Full Text Request | Related items |
