7S globin(β-Conglycinin)is one of the main components of soybean seed storage protein.It is trimeric glycoprotein composed of α-(5758KD),α’-(5772KD)and β-(4552KD)subunits,all above three subunits are the main allergens of soybean.The current study shows that 7S globulin subunit deletion mutants can remove the sensitized protein,at the same time,improve the nutrition and processing quality of soybean protein,but the role of each subunits on the soybean seed protein nutrition and processing quality and contribution size has no definite conclusion,The molecular mechanism has not yet been resolved.So far,there has been no reports of new soybean materials of α’-subunit deletionwith genetic background of China,and the genetic effect of singleα’-subunit deletion is not clear.In this study,we used the backcross method to obtain the new soybean lines of α’-subunit deletion and the near isogenic line with the genetic background of China,and make a comprehensive evaluation of its main agronomic traits,nutrition and processing quality.Clarify genetic effects of α’-subunit deletion;the α’-subunit deletion gene was preliminarily located,which laid the foundation for molecular marker assisted breeding and map cloning and functional verification of the gene.The results are as follows:(1)Using backcross breeding method obtained 12 soybean new lines with low susceptibility protein α’-subunit deletion in genetic background of China:C0909,C4802,C4808,C4809,C4819,C4821,C4833,C4840,C5206,C5210,C5211,Cc5-7 and all lines were α’-subunits deletion.Among them,in the agronomic traits,the yield of four plants was significantly higher than their parent Dongnong47;in the nutritional quality,The content of oil was significantly higher than their parent Dongnong47,the contents of 17 amino acids were increased and the free arginine content increased,free alanine,free valine,free methionine,free benzene Acid,free histidine and other five kinds of free amino acid components were significantly increased;in the processing quality,tofu products,gel hardness and tofu protein concentration increased significantly,viscosity and other texture characteristics were significantly improved.(2)using Dongnong47 as reincarnation parent,through the 6 generation backcross,4generation selfing(BC6F4),one groups of near isogenic lines were constructed,and named as NIL-Δα’,genetic analysis results show that: single α’-subunit deletion increased plant height and yield of near isogenic lines.In the nutritional quality,The content of 17 amino acids and free amino acid showing a different degree of rising trend.In the processing quality,the gel hardness of tofu products increased significantly,the dry weight of tofu and the plasma concentration of tofu were significantly increased,and the texture characteristics such as elasticity and chewiness were significant improved.(3)The α’-subunit deletion gene was preliminarily located,and the gene pool was constructed by BSA(Bulked segregant analysis)method.Using SSR(Simple Sequence Repeat)markeranalysis,mapmaker3.0 software make map,The α’-subunit deletion gene(cgy-1)was preliminarily located on the chromosome No.11(chromosome 1),and the α’-subunit deletion gene(cgy-1)was initially mapped to soybean chromosome 10(O chain group),between Sat307 and Sat231,and the distance from Sat307 about 6.1cM.
