Indigofera amblyanta is a good plant for soil and water conservation,which is more and more widely used in the garden slope protection project,but its seed is not durable storage,the seed vigor declined rapidly with the increase of storage time.In order to prolong the storage life of seeds of Indigofera amblyantha,this article studied the changes of seed vigor with the increase of storage time under different storage conditions,so as to provide the technical basis for the safe storage of Indigofera amblyantha.According to the climate of the field test site,the storage conditions were set up as 2%,6%,10%moisture content and-10 centigrade,5 centigrade and room temperature combined storage.Then the experiment studied the changes of germination rate,field emergence rate,electrical conductivity,soluble sugar content and malondialdehyde(MDA)content under different storage conditions in different seasons,and through the analysis of variance and linear regression of the experimental data of Indigofera amblyantha seeds during storage of seed vigor,internal physiological and biochemical changes were analyzed and summarized.The results are as follows:(1)the standard germination rate and field emergence rate of the seeds were decreased with the increase of storage time.The standard germination rate during the storage period of 3 months,6%water content storage can maintain high germination rate under different storage temperature,longer storage time,the lower the temperature,the lower seed moisture content,germination rate decreased more slowly.The field emergence rate was highest under the condition of 5 centigrade and water content of 6%.Both of them were the fastest descent rate at room temperature and 10%water content.Germination rate to December,down to 0,the rate of field emergence to September,down to 3%.(2)The low water content and low temperature storage could slow down the physiological and biochemical rate of the seeds,but there were some differences in the changes of the indexes under different storage conditions.The conductivity of leachate increased with the increase of storage time.When the storage temperature was-10 centigrade or 2%moisture content,the conductivity increased slowly,and the conductivity increased rapidly at room temperature and 10%water content.The soluble sugar content with prolonged storage time,increased after the first drop,the effect of moisture content and storage temperature to the change of soluble sugar content is not obvious.MD A content increased gradually with the storage time,and it was the slowest in the storage of-10 centigrade and 2%water content,and rise rapidly at room temperature or 10%moisture storage.(3)The effects of storage conditions on seed germination and physiological and biochemical indexes(except for the storage period of MDA to December)were significantly different(P<0.05).The effects of storage conditions on germination rate and field emergence were very significant during storage.The effects of storage conditions on the electrical conductivity were more and more significant with the storage time prolonging.The effects of storage conditions on the soluble sugar content and electrical conductivity were not significantly different with the storage time.(4)The germination rate and field emergence rate showed a significant positive linear correlation,the correlation coefficients were greater than 0.9under different storage conditions.There was a negative correlation between conductivity,MDA content and germination rate,and the linear relationship between conductivity and germination rate was strong.Under different storage conditions,the absolute value of correlation coefficient is greater than 0.85.The soluble sugar content could not clearly reflect the seed vigor,and there was no significant relationship between soluble sugar content and germination rate.(5)-10 centigrade,2%moisture content and 6%moisture content storage conditions had the best effect on slowinglndigofera amblyantha seed deterioration.(10%moisture conten,at room temperature)storage conditions of Indigofera amblyantha seed deterioration most rapidly,the seed storage life of less than 1 years.
