| Ceftiofur is the third generation cephalosporin that veterinarians use specially,which was developed successfully by Pharmacia & Upjohn Company in the 1980 s.As the third generation cephalosporin,it can inhibit the synthesis of the cell wall and then lead to the death of bacteria.That is the antibacterial mechanism of ceftiofur.The effect of ceftiofur on streptococcus is greater than that of fluoroquinolones.The antibacterial activity of it is stronger than that of principen.Because of the wide antibacterial,the ability of inhibiting and the great killing pathogenic microorganisms,the side effect is little and the residue is low,it has been approved and accepted by more and more people,since ceftiofur was produced.In addition,it was approved for the treatment of bacterial diseases in animals.Since 1988,Ceftiofur sodium has been formally approved by North America,and then the treatment of respiratory diseases in pigs,sheep,cows,beef cattle,horses and the prevention and cure of infectious diseases of pets was approved by some European countries and Japan one after another.Currently,livestock breeding is growing in scale in our country.The breeding density is large and the environment is bad,which lead to epidemic and outbreak of infectious diseases easily for the livestock.As a result,the epidemic situation could appear on a large scale.What’s worse,a number of livestock could die,which can cause an enormous loss to the stockbreeding.As a new kind of antibiotic for animals,ceftiofur sodium can solve problems that bacterial diseases bring about in poultry farms to some extent.With the successful synthesis and mass production of ceftiofur sodium by domestic enterprises,The cost of the synthetic materials is getting lower and lower,and the application of ceftiofur sodium in veterinary medicine is more and more extensive.At the same time,in order to make it clear that if ceftiofur sodium can be applied scientifically,reasonably and safely in different kinds of livestock and poultry,we promoted the in-depth study of the drug in a variety of animal body inside and outside.By detecting the concentrations of ceftiofur sodium in the partridge chicken by High Performance Liquid and studying the ceftiofur sodium pharmacokinetic behavior in the body of partridge chickens,we can judge and understand clearly the ADME process of ceftiofur sodium in the body of the chicken(i.e.absorption,distribution,metabolism and excretion process).At the same time,we can ascertain ceftiofur sodium administration routes and timemore scientifically and establish the research method of pharmacokinetics and tissue distribution of determination of ceftiofur sodium.Thus we can provide reference for the safe and rational drug use clinically and for the establishment of the drug with drawal period and the prevention of drug residues.We used two ways of drug delivery,oral and intravenous injection.We collected venous blood under chicken wings at different times after administration.The HPLC method was used to detect and analyse the prototype drug of ceftiofur sodium by plasma samples and tissue samples.After that,we can study the pharmacokinetics characteristics of ceftiofur sodium and analyse the bioavailability of ceftiofur sodium in the body of chickens.By the study of pharmacokinetics of ceftiofur sodium in the body of chickens,we found that the pharmacokinetic parameters of 20 mg/kg that the chickens were treated with intramuscular injection ceftiofur sodium were showed as follows.The Cmax of peak plasma concentration is(6.337 ± 3.29)μg/m L.The Tmax of peak time is(0.25 ± 0.00)h.The elimination half-life t1/2 is(0.851 ± 0.23)h.The elimination rate constant,Ke is(0.883±0.32)h-1,AUC0-t is(5.338 ± 2.85)μg·h·mL-1,AUC0→∞ is(5.417 ± 2.87)μg·h·mL-1,the mean residence time is(0.837 ± 0.07)h,CLtot is(72.950 ± 28.53)m L·min-1·kg-1,the apparent distribution volume Vd is(5.250 ± 2.20)L·kg-1.The prototype drug of ceftiofur sodium was not detected in the tissue after giving ceftiofur sodium to the chicken by intramuscular injection.By studying the pharmacokinetics after giving ceftiofur sodium of 20 mg/kg to the chicken by oral,the prototype drug of ceftiofur sodium was not detected in blood and tissues of the chicken,which shows that ceftiofur sodium can’t be absorbed by oral. |
