Banana (Musa SPP.) is a large perennial evergreen endogen herb, native to tropical and subtropical regions, is one of the main fruit in the world, and is also an important food and cash crops. Drought is always a worldwide problem,seriously affected the yield and quality of the banana, the banana production is one of the major abiotic stress factors.miRNAs is a kind of coding protein,has open reading frame of the endogenous stem ring single-stranded RNA structure,widely exists in eukaryotic organisms,and closely related to growth and development. At present, the existing various plants under abiotic stress related miRNA research reports, but for banana miRNA research under drought stress were not reported. This study from the early stage of the research for high-throughput sequencing 60 drought related micrornas, on the basis of article 39 of the selected (35 known,4 unknown). Brazil banana seedlings as material,to simulate the natural biological process of drought stress, banana to a domesticated by the method of real time fluorescence quantitative drought (drought) directly, and after the domestication of drought under two kinds of processing the miRNA expression differences analysis, for the further study of miRNAs on the regulation mechanism of banana drought stress response laid a foundation. This study’s main findings are as follows:(1) Through the stem loop primers rt-pcr method, verified the drought related miRNA expression in banana leaves. Of 39 banana drought miRNAs expression analysis results show that the majority of miRNAs in 10 point (0,2,4,6,8,10,12,14,16,18 days) of two kinds of drought treatment (drought DHN directly, after the domestication drought DH) were detected with expression, and in the expression of two under drought treatment has obvious scheduling. Part of miRNAs in different treatment,different time point expression was detected. Further expression analysis results show that the article 39 of micrornas in both under drought treatment in banana leaf expression showed obvious difference. Article 17 the micrornas in domesticated under drought treatment degree is higher than the average response after the domestication degree of average response under drought treatment.Directly under drought treatment, least miR396c - 5 p response, the highest degree in miR397a response; Domestication under drought treatment, least miR394 response,miR399a response to the highest degree. Response to drought strongly micrornas may belong to the direct control of banana drought resistance mechanism of micrornas, further research will be banana drought-resistant related micrornas main object.(2) The miRNA expression pattern analysis under different drought treatment results show that the miRNA has different degrees of drought stress response. Directly under drought treatment, micrornas in 10 days most violent response to drought; Domesticated under drought treatment, micrornas in 8 days most violent response to drought. From the point of view of reaction time, the domestication of banana plants, in the face of the drought stress of micrornas in plant response to earlier than without drought domestication of the banana plant, its mechanism may be similar to the immune response, when drought house-trained plant again face drought micrornas will more rapid response earlier.(3) Using qPCR target gene technology for online prediction of six representative miRNA family and two unnamed six of the miRNA target genes are verified. The results showed that most of the expression of target genes and expression of corresponding miRNAO to 10 days appear the phenomenon of negative correlation,as it happens and the miRNA degradation or translation through inhibiting the mechanism of action of target genes, preliminary verify the authenticity of micrornas predicted target genes. |
