Retained Fetal Membarances?RFM?was defined as the fatal membrance was not discharged from the uterine of a cow within postpartum 12 h.There was a high incidence in RFM,secondary diseases such as mastitis,laminitis,endometritis would happened on the cow with RFM.Feeding DCAD with Ca2+ can reduce the risk of RFM,Ca2+ has mediated many process as second messengers,such as the concentration of muscle,the release of hormone and inflammation.The metabolism of Ca2+ in RFM was study in this experiment to lay the foundation of RFM and find the effective method of treatment for RFM.12 dairy cows with the similar of weight,age,fetal times and health condition were selected.It was divided of two groups,RFM and control,with average of 6 cows,respectively.The plasma of cows with RFM and without RFM were collected during at antepartum 25 days and postpartum 6,12,24 h.Then concentration of Ca2+ in plasma simples were detected,and the data was analyzed by SPSS19.0.The m RNA expressions of the proteins with the function of promoting Ca2+ get in the cells?TRPC1,TPPV1,S100A1,SERCA,IP3R1,CACNA1 D,Ry R1,PLC??,the m RNA expression of the proteins with the function of promoting Ca2+ get out of the cells?PMCA,SLC8A1?and m RNA expressions of the protein with the function of storing Ca2+?CASQ1,CALM?were detected in RFM group and control group by Q-PCR.The expression of some proteins were detected by Western blot?PMCA,PLC?.The m RNA expressions of the proteins with the function of regulated the contraction of smooth muscle?MLCK,MLC,Rho,Rho A?were detected in RFM group and control group by Q-PCR.The results showed that the concentrations of Ca2+ have no significantly differences between with RFM and without RFM of the cows during 25 days at antepartum in plasma?P>0.05?,however,the Ca2+ concentrations of plasma of cows with RFM were significantly lower than control group within antepartum 1 days?P<0.05?.The Ca2+ concentrations of plasma of cows with RFM were significantly lower than control group at postpartum 6 h,12 h,24 h?P<0.01?.The m RNA expressions of the proteins with the function of promoting Ca2+ get in the cells?TPPV1,S100A1,SERCA,Ry R1,PLC??were much lower significantly?P<0.01?than the m RNA expression in control group,the m RNA expressions in RFM of TRPC1,IP3R1 and CACNA1D?P<0.05?were lower significantly than the m RNA expressions in control group.The m RNA expression of the proteins with the function of promoting Ca2+ get out of the cells PMCA was much higher significantly?P<0.01?than the m RNA expression in control group.The m RNA expression in RFM of SLC8A1?P<0.05?was higher significantly than the m RNA expression in control group.The results of Western blot from PMCA and PLC? were the same as the result of Q-PCR.The results displayed that the m RNA expressiosn in RFM of MLC?P<0.01?were much lower significantly?P<0.01?than the m RNA expression in control group.The m RNA expression in RFM of MLCK?Rho A ?Rho were lower significantly?P<0.05?than the m RNA expression in control group.The results of this experiment suggested us,the m RNA expression of proteins which regulated the Ca2+ get in and out the cells and the proteins which regulate the contraction of smooth muscle in RFM group were significantly different from control group.They participated in the progress of the fatal membrances get off from placentas of cows and played an important role in RFM by regulated the smooth muscle of vascular in placenta of cows.
