SCFA provides not only to their involvement as the major source of nutrients in ruminants, but also serve as regulator of signal transduction.The study was isolated the rumianal smooth muscle cells and epithelium cells of goats in different ages by the methods of collagen and enzyme digestion respectivety. The study detect the protein and mRNA change of MYL9?MLCK in the Ca2+ dependant pathway, ROCK1?ROCK2? RhoA in the ROCK/RhoA pathway and PKC??PKC? in the PCK pathway that in the co-culture rumianal smooth muscle cells with epithelium cells which be incubated with SCFAs.The study aimed to evaluate the effect of SCFAs to rumianal muscle contraction by researched the mechanism of smooth muscle contraction.The results are as follows:1. There aren't significant difference among the four groups about the expression quantity of MYL9 gene in translation level (p>0.05),and SCFAs down regulation the MYL9 gene in transcripitional level.SCFAs up regulation the expression of MLCK proptein and mRNA.There are significant difference in the rumianal smooth muscle cells of goat in different ages about the expression of MYL9 and MYLK gene (p<0.01)2. In the translation level,ROCK1 and RhoA expressed stably among the groups,but there are significant difference among the groups about the expression of ROCK1?ROCK2?RhoA mRNA(p< 0.01),especially the propiona group.And there are significant difference in the rumianal smooth muscle cells of goat in different ages about the expression of the revelant gene and protein in the ROCK/RhoA pathway (p< 0.01).3. The PKC8 protein expressed stably among the groups,the PKC? protein opposite (p< 0.01),SCFAs up regulation its'expression.However,SCFAs groups down regulation the expression of PKC? and PKC? gene in the transcripitional level compare with control groups..There are significant difference in the rumianal smooth muscle cells of goat in different ages about the expression of PKC? and PKC? gene (p<0.01)In conclusion, in transcripitional level,SCFAs down regulation the MYL9?PKC8 and PKC? gene,and it up regulation the MYLK?ROCK1?ROCK2?RhoA gene.SCFAs also up regulation the MYLK and PKC? in translation level.In the process of rumnial development from goat,Ca2+ dependant pathway?ROCK/RhoA pathway and PKC pathway all have regulation effect on rumnial muscle contration.
