Manganese(Mn)is widely distributed in the nature of an element,manganese show a strong biological function,and the synthesis of enzyme,three substance metabolism,hematopoiesis,immune system and reproductive function has a direct relationship.Manganese is the body essential nutrients,but excessive manganese is harmful to organisms.Manganese damage to nerve,reproductive and immune system has been confirmed.But manganese on immune system damage research is still very rare birds.However,the effect of Mn-induced on in chicken immune organs is still poorly understood.This study was conducted to investigate the toxic effect of Mn on chicken immune organs.7-day-old 180 chickens were fed either a commercial diet or three Mn Cl2-added commercial diets containing 600,900,and 1800 mg Mn/kg,respectively.On the 30 th,60th,and 90 th day of the experiment,bursa of Fabricius(BF),thymus,and spleen were collected,respectively,and then the m RNA levels of IL-4,IL-7,IL-10,IL-17,TGF-?4,NF-?B,TNF-?,COX-2,PGE2,iNOS and IFN-? were examined.And using Multiple correlation,principal component analysis(PCA)and R cluster analysis these statistics analysis method to comprehensive data processing,more intuitive response under the condition of the chronic manganese poisoning,animal immune organs within IL-4,IL-7,IL-10,IL-17,TGF-?4 and IFN-? mRNA content and manganese content has a close contact.Enrich the contents of the immune toxicology research.Conclusion after discussion:1.30 and 60 days Manganese poisoning raised NO Content,iNOS Vitalite,IL-4,IL-7,IL-10,IL-17,TGF-?4,NF-?B,TNF-?,COX-2,PGE2 and iNOS mRNA expression and Inhibition of IFN-? m RNA expression in BF,thymus,and spleen.The immune organ responds by regulating the expression of its cytokines and inflammatory factors.Manganese poisoning decreased secretion of all detection factor,in BF,thymus and spleen in 90 days.Manganese poisoning cause injury immune and inflammatory Injury on morphology in BF,thymus and spleen.There were present in the dose effect in 60 days of the thymine IL-7,60 days of the spleen NF-?B,30 days of the thymus iNOS and 30 days of the spleen PGE2 mRNA expression.2.IL-4,IL-7,IL-10 and IL-17 mRNAexpression are close relationship,and into a significant negative correlation with IFN-?.Manganese poisoning cause immune injury and inflammatory Injury in BF,thymus and spleen.
