The Correlation Research Of Ginseng Saponin Content With Saponin's Biosynthesis Key Enzyme Activity And Gene Expression

Ginseng,as authentic medicinal materials in China,the main active ingredients of it is ginseng saponin.At present,more and more researchers pay attention on the synthesis pathway of ginseng saponin.However,the correlation of enzyme activity and gene expression of key enzymes in saponin synthesis pathway has not been reported yet.It has important theoretical and practical application value to investigate the relationship for the ginseng'sidentificationand traceability andthe use of biotechnology to improve ginseng saponin content.The seven key enzymes were studied which are in the main way of the synthesis ofdammarane-type ginsenosides.The correlation between the key enzyme's activity?gene expression and saponins was been analyzedin different regionsand years.The enzymes that has the regional or year's characteristics have been discovered..In this study,the fresh ginseng samples' s hairy roots in different years and region in JingYu county,FuSong county,Wangqing county of Jilin province were as the material.The spectrophotometry and HPLC method were used to detect total saponins and monomer saponins.The method of q-PCR method was used todetect gene expression the key enzymes(HMGR,FPS,SS,SQE,DS,P450,GT).Using colorimetric method,,ELISA and HPLC and LC-MS and GC-MS method to detect the seven key enzyme activity.Correlation results is analyzed by using SPSS software combined with R language.This topic research discovered that the monomer saponins in different years has significant correlation with total saponins of ginseng saponin andthe content of Re in different regionshas great significance traceability.The DS and GT's enzyme activity and gene expression in ginseng has great significance in the accumulation of saponin in different years.Th HMGR and DS's gene expression will have important reference value in traceability.The Research of this paper provide the support of biochemistry and molecular biologyin-depth study of ginseng saponin biosynthesis pathway.Meanwhile,the research provides a theoretical basis in the year of ginseng's identification and the establishment of regional traceability index.
