Study On The Pharmacognosy Of Caulis Clematidis Armandii And Its Substiutes

Objective:Chuan Mutong is the dried cane of Clematis armandii Franch.or Clematis montana Buch.-Ham.of Ranunculaceae.Chuan Mutong has the effects of diuresis,treating stranguria,clearing away the heart-fire,relieving restlessness,restoring menstrual flow and promoting lactation.Now,the market mixed-use is severe.It is hard to identify prepared herbal medicine in small pieces ready for decoction of Chuan Mutong,so the medication safety and effects cannot be guaranteed.The research aimed to explore the pharmacognosy characteristics,microscopy,thin layers qualitative determination and DNA bar codes as well as gene and chemical double-fingerprint research of HPLC feature pattern so as to form a comprehensive study system.Meanwhile,the initial research on chemical constituents and diuresis effects of Clematis argentilucida.W.T.Wang and Clematis gouriana Roxb.enjoying higher market share is conducted in order to provide support for reasonable use of Chuan Mutong.Method:In this research,with application of paraffin embedding technique and saffron-fast green staining,the stem of Chuan Mutong are systematically identified,and the powder of four kinds of medicinal materials were compared.According to medicinal materials in the control group,the TLC identification conditions of Chuan Mutong are established.Examination items like water content,total ash and extract of medicinal materials of Chuan Mutong are tested.With adoption of PCR method,ITS2 sequence is amplified into DNA in medicinal materials and prepared herbal medicine in small pieces ready for decoction and the ITS2 sequence of the sample is comparatively analyzed through software.HPLC specific chromatogram of Chuan Mutong is established.With utilization of"Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine",similarity comparison is conducted between collected samples and Clematis armandii Franch.and Clematis montana Buch.-Ham.Finally,pesticide effect of the statutory breed of pharmacopeia and breed habitually used in the market were distinguished in the paper and the diuretic efficacy of pharmacopeia and non-pharmacopeia breeds were initially compared in paper.With application of self-made metabolic cage,comparative research is conducted on diuresis effects of Clematis armandii Franch.,Clematis montana Buch.-Ham.,C.argentilucida.W.T.Wang and Clematis gouriana Roxb..Result:The results of micrograph showed,primary rays of Clematis armandii Franch.are 17 while those of the others are 12;the thick-walled cell band in the phloem of Clematis montana Buch.-Ham.do not form circles in continuity and interrupts at the pith rays;Clematis argentilucida.W.T.Wang presents non-obvious six grooves whose catheter holes are large;Clematis gouriana Roxb.appear to be petal-shaped.The results of powder showed,phloem ray cells,stone cell and vessels of Clematis armandii Franch.,Clematis montana Buch.-Ham.,Clematis argentilucida.W.T.Wang and Clematis gouriana Roxb.have differences in shape and size.The results of based on the ITS2 DNA barcode showed,Clematis fargentilucida.W.T.Wang occupies the principal place in the prepared herbal medicine in small pieces ready for decoction.Meanwhile,as regulated by pharmacopoeia,the use of Clematis armandii Franch.is less without existence of Clematis montana Buch.-Ham.The results from NJ tree showed,the relationship among Clematis montana Buch.-Ham.,Clematis argentilucida.W.T.Wang and Clematis gouriana Roxb.are close.According to HPLC specific chromatogram comparison,similarity in chemical components of Clematis argentilucida.W.T.Wang,Clematis gouriana Roxb.and Clematis montana Buch.-Ham.is extremely high;through research on diuresis effects,it can be indicated that Clematis argentilucida.W.T.Wang has obvious diuresis effects.Conclusion:1.This study established a systematic identification method,can effectively distinguish between Chuan Mutong and plants of the genus Clematis(Ranunculaceae).2.This study verifies Clematis argentilucida.W.T.Wang and Clematis gouriana Roxb.replace also the rationality of the use.Thus,Clematis argentilucida.W.T.Wang is recommended to be classified into Chinese Pharmacopoeia.
