Human granulocytic anaplasmosis(HGA)is a kind of zoonotic disease caused by neutrophil phagocytophilum(AP)infecting human peripheral blood neutrophils.It is a natural epidemic disease..Aphids are the main vector of the disease,such as the whole ditch hard clam,the forest leather tadpole,the shoulder blade hard palate,and the Pacific hard palate.HGA is mainly transmitted by infecting different host animals through mites,which are widely used to infect humans,livestock,rodents and various wild animals.The persistent infection of various wild animals is to maintain the long-term presence of this disease in nature.Important conditions.In recent years,there have been more and more cases of human and various animal infections of phagocytic anaplasma in China and abroad.In China,phagocytic anaplasms have been detected from various animals,baboons and humans,indicating the infection of anaplasmosis.The number and distribution range are gradually expanding.Xinjiang is located in the hinterland of Asia-Europe continent.It has a vast territory and diverse ecological environment.Sheep farming is a traditional animal husbandry in Xinjiang.Sheep are closely related to various cockroaches and rodents,which greatly increases the infection of human and animal granulocytic anaplasma.risks of.We conducted a molecular epidemiological investigation on the anaplasm of sheep infected with different species in the northern foothills of the Tianshan Mountains in Xinjiang,pathological observation and animal infection experiments to determine the infection rate of different species of sheep anamorpha,genetic characteristics and pathogenicity of the pathogen.The health of the development is escorted.The main research methods and results are as follows:(1)Infectious epidemiological survey of three breeds of sheep in different regions of Xinjiang: 300 samples of Kazakh sheep from Musi Township,Nileke County,Xinjiang,China,and 300 blood samples from Manas Lake,Manas Guangdong Township 300 Suffolk sheep blood samples were tested for invisible 16 s RNA by nested PCR.The results showed that the positive rate of Ili Kazakh sheep invisible was 28%,and the Manas Lake sheep invisible body positive rate was 17.7%.Manas Suffolk The positive rate of gram sheep invisible was 24%,and the overall infection rate was 23.3%.Among them,the infection rate of Kazakh sheep in Musi Township of Yili Nileke County is the highest,and that of Manas County is the lowest.(2)Genetic evolution analysis of Xinjiang sheep invisible 16sRNA: 30 positive samples were randomly selected and amplified by 16 s RNA specific primers.After sequencing,5 differential sequences were selected for genetic evolution tree analysis.These sequences were named Sheepp.,Sheep2,Sheep3,Sheep4,Sheep5.DNAstar was used to compare the above gene sequences with the typical in vivo sequences in GenBank.The results showed that the sequence homology between sheep1-5 was over 88.8%.The five positive sequences have higher homology with Jilin,Zhejiang and Heilongjiang,all of which are above 91.6%.The homology between sheep1 and Zhejiang source is as high as 98.2%.The partial sequence homology with Japan,Russia and the United States is in the same sequence.More than 91.6%,the homology with Russian-derived anaplasm is as high as 99.2%.(3)Invisible pathogens artificially infected sheep and mice: Blood samples from sheep that were positive by invisible PCR were used to make blood smears and observe the pathological characteristics.The sheep whose test was positive by PCR were inoculated intraperitoneally into the sheep which were negative by the invisible body.The mental state of the experimental animals was observed every 2 days,the jugular vein was collected and detected by PCR,and the experimental group B was detected on the 8th day of the experiment.One sheep in the group had positive PCR,but it disappeared positively in the subsequent blood sampling PCR test.The blood of the sheep positive by PCR was inoculated into the negative mice by abdominal cavity,and the mental state of the mice was observed every 2 days.The mouse spleen was detected by PCR and the mice were not infected with the anaplasm.After the 12 th day of the experiment,the remaining mice were dissected and the infected spleen was obviously swollen.In summary,different breeds of sheep in Xinjiang have anaplasmic infections and high infection rates;anaplasms cannot infect sheep and mice through intraperitoneal vaccination,and their transmission pathways may have to be transmitted through vector sputum.
