Theileria's disease of ovine is a protozoan disease of the blood transmitted by ticks,The disease infection caused by animal fever,Superficial lymph node enlargement,anemia,weight loss and other symptoms,severe cases can cause death.Superficial lymph node enlargement,anemia,weight loss and other symptoms,severe cases can cause death.The domestic and foreign related Neitaile ovine blood disease epidemic research and detection methods more,But for the ovine in the south of Xinjiang theileria's disease infection and pathogens have been reported less.The purpose of this study is to examine the infection and PCR technology to identify the areas in the application of Theileria ovine blood microscope,And identification of its pathogen,In order to provide scientific basis for diagnosis and prevention of anaplasmosis in ovine industry.1.In order to know the prevalence of theileria's disease in ovine in kashgar area,From November 2015 to January 2016,Blood samples were used to examine the blood samples of 258 local adult sheep in 11 kashgar area.The results showed that the theileria sample was 151 and the total infection rate was 59.0%.The infection rates of theileria r insects in different sampling points were different.Among them,the infection rate of theileria's insects in Shache ovine was the highest,75%,while that of kashgar ovine was lower than,which was 8.3%.The survey results show that kashgar ovine Taylor disease epidemic,should strengthen the comprehensive prevention and treatment of the disease.2.For the situation and the types of infection in southern Xinjiang to further grasp the ovine theileria.theileria based on universal primers,to detect the region of 637 ovine blood samples by PCR method DNA,400 positive samples of theileria were found,with a total infection rate of 62.8%.Taylor from different sampling points of ovine infection rate differences,among them,Shache,Yengisar and the theileria infection rate of Zepu ovine were as high as 100%,Shaya ovine theileria infection rate is low,4.0%.With ovine theileria,especially Taylor and Lu theileria insect specific primers of more than 400 copies of theileria positive samples,1 samples(sample No.1443)on site primers were identified as positive,but in 3 specific primers were amplified bands.3.A random sample of 56 ovine positive samples and 1443 samples were sequenced by PCR,Successful55 ovine theileria sequences and 1443 sample sequences.Blast than in NCBI,using Clustal X 2.11 software were compared and sequence.All ovine found consistent Taylor worm sequence from Iran ovine andovine theileria(Gen Bank accession number KX273858),Turkey ovine theileria(KU342701),Chinese ovine theileria(FJ603460)sequence homology was 100%,identified as sheep theileria.The sequence of 1443 samples from Iran cattle ring theileria(KX273857),India cattle ring Taylor(KT736498),the insect Chinese Sika Deer ring theileria(KT959231)sequence homology was 100%,identified as theileria ring.
