Abstract/Summary:
Theileria is an obligatory intraerythrocytic protozoan parasite transmitted by ticks and it causes economic losses to the livestock industry worldwide. Nucleotide anabolism metabolism pathway of two Theileria sp.(T. luwenshuni and T. uilenbergi) was analyzed with bioinformatics. oprt gene was cloned and expressed, and an animal protective trial of recombinant protein r OPRT was performed in control sheep group and experimental sheep group sheep. The main contents showed below.1. Nucleotide metabolism pathway of two Theileria spp.(T. luwenshuni and T. uilenbergi) was analyzed. The results showed that there were five proteins related to nucleotide metabolism via COG analysis. Of them, there were four proteins participating in the beginning way, only one protein participating in the remedy way. And orotate p Hosp Horibosyltransferase and adenylate kinase were critical enzymes as beginning synthesis and remedy synthesis of nucleotide, respectively.2. The functional fragment of oprt gene of T. uilenbergi was cloned and expressed, and the reactionogenicity of the recombinant protein was analyzed. The results showed that the purified OPRT protein was inclusion protein, and there was cross reaction with the two positive sera of T. luwenshuni and T. ovis, respectively, which indicated that the OPRT homogogous gene was also existed in the these two Theileria spp.3. Animal protection test was taken up for further investigation of OPRT as the candidate antigen.Statistical analysis showed that fever of sheep lasted longer in control group than in immunized group,moreover, an obviously shorter Theileria-carrier period was observed in immuneized group via blood smear. In control group, high infection rate of theileria was at 25、26、29 day, however, the time was at26、32、33 day in immune group, which was obviously. Determination of hemoglobin content(Hb)showed a significant decrease in control group and a slightly decrease in immunized group. The minimum value in control group was smaller than in immunized group and appeared earlier. Blood sampling using cell counter showed red blood count along with clinical symptoms showed that severe period of anemia was tediously long in control group.4. Totally, 2351 blood samples collected from goats and sheeps aged 8 to 20 months from 47 regions of 21 province between 2012(May) and 2013(June) was detected with indirect ELISA to acquire epidemic status of Theileria in China. Results indicated prevalence in Gansu province was highest with the prevalence of 76%, and the prevalence was lowest with 0% in Hebei province, in which there was no detection of Theileria.
