| Volvariella volvacea originated in China,is an important member of edible fungus and called Chinese mushroom.Volvariella volvacea is loved by people.As a high temperature fungi,Volvariella volvacea mainly plants in summer.At present,Volvariella volvacea production mainly uses a cultivation method of solid inoculation fermentation material.But a variety of reasons lead to lower yield of Volvariella volvacea production,such as the quality of open fermentation quality is difficult to control and production stability is poor,and cell age of solid spawn is inconsistent.So the improvement of Volvariella volvacea cultivation technology is particularly important.This experiment starts from the formula and cultivation methods,trying to use liquid spawn instead of solid strain and optimizing the Volvariella volvacea liquid spawn culture medium and culture condition.In addition,using clinker cultivation instead of fermentation cultivation.Conclusions are as follows:(1)By ITS molecular sequencing combined with morphological characteristics,strain YL-93 which provides by the laboratory is Volvariella volvacea.(2)According to the growth of mycelium,screening the most suitable mother culture medium for V volvacea YL-93 was P5 and the recipe was peeled potato 200g/L,bran 50g/L,glucose 20g/L,agar powder 20g/L and pH 7.5.The best solid spawn culture medium for V.volvaceaYL-93 was screened and the recipe was wheat grain 98%,calcium sulfate 2%.(3)Through single factor test and orthogonal test,optimum liquid spawn culture medium for V.volvacea YL-93 was determined.The best liquid spawn culture medium formula was glucose 15g/L,yeast extract 6g/L,MgSO4·7H2O 1.5g/L,KH2PO4 0.5g/L.The culture conditions of V.volvaceaYL-93 liquid spawn were optimized.The optimum conditions were inoculum concentration 5%(v/v),the initial pH 7~7.5 and the volume of liquid 75~100mL for each 250mL triangular flask,shaking culture for 60 hours protected from light at 30 degrees Celsius.The number of effective pellet can reach 45 per milliliter and the dry weight of mycelium reached 5mg per milliliter.(4)In the clinker cultivation experiment,under the same conditions,the time of liquid in oculation of mycelium was about 9 days,which was shorter than solid inoculation.At the same time,the formation time of the liquid inoculation primordia was about 12 days,which was shorter than 19 days of solid inoculation.Finally,liquid inoculation yield increased by 28.6%compared with solid inoculation.(5)By V Volvacea YL-93 fresh yield for each edible fungi bag as the index,The optimum proportion of straw and corn straw was selected by the clinker cultivation experiment.The optimum ratio of raw material was crushed corn straw 44%,crushed rice straw 44%,wheat bran 10%,CaSO4 2%.Using response surface method to determined the optimum conditions of V.Volvacea clinker cultivation,that is,cultivation material moisture 71.3%,soaked calcium hydroxide solution mass fraction 3.2%,liquid spawn inoculum concentration is 4.8%.Under this condition,V.Volvacea YL-93 theoretical yield is 22.47g/bag.Through the batch verification test,the average yield of V Volvacea YL-93 is 21.85g/bag. |
