Ganoderma lingzhi is a kind of medicinal material which has broad application prospect.As one of the key ingredients in the Ganoderma lingzhi,Ganoderma triterpenoids has important pharmacological actions,however,its content in G.lingzhi cell and the production yield were low,which limits its large-scale application.Therefore,this thesis aims to study the effects of exogenous elicitors(coix seed oil)on the biosyntheisis of G.lingzhi triterpenoids during the submerged fermentation of G.lingzhi to find the suitable concentration of coix seed oil,which can significantly increase the synthesis of triterpenoids’ and production yield,and further to analyze the basic signal transduction mechanism of NO and H2O2 involved in the synthesis of G.lingzhi triterpenoids by coix seed oil.The main results are as follows:1.The elicitor(coix seed oil)could significantly promote the mycelial growth and the triterpenoids synthesis at the concentration of 1.5g/L.Elicitors are most sensitive when add it to hyphal cells in the stages of end of growth to early stabilization,this kind of indecement can induce G.lucidum mycelia produce secondary metabolite in a long time.2.Coix seed oil could induce effectively the improvement of the yeild of NO,PAL’s activation and the content of CYP450 in G.lucidum cells.NO can play a role in PAL’s activation and the produce of CYP450 as a free signal molecule,it is a signal transduction in G.lucidum cells,induced by coix seed oil of PAL’s activation and the produce of CYP450,NO will be also produced as the signal molecule in the compound of G.lucidum triterpenoids,but it is not the only one.3.H2O2 is located in the PAL’s activation and the produce of CYP450 upstream.The pathway of signal transduction mediated by H2O2 was induced by coix seed oil,but the H2O2 pathway was not the only way to induce the synthesis of G.lucidum triterpenoids by coix seed oil.4.Both NO and H2O2 joined the process of signal transduction by a contrast experiment,compounding the G lucidum triterpenoids induced by coix seed oil,NO and H2O2 as signal molecule can cooperate in the process of coix seed oil induced.In addition,in the two comparison group of SNP+H2O2+DMTU+IMD and SNP+H2O2+cPITO+L-NNA,PAL’s activation and the content of CYP450 are both higher than H2O’s.It is indicated that H2O2 and NO could be signal molecule to mediate the coix seed oil’s inductive effect solely.
