Cadmium is one of the t oxic heavy metals.Trace cadmium exposure can cause great damage to the health of animal and human.Cadmium can cause damage of chicken testis and kidney.This experiment was designed to investigate the protective effects of Ganoderma triterpenoids on cadmium-induced chicken testis and kidney injury.Eighty healthy 7-day-old Hyland white chickens were randomly divided into four groups.The cadmium poisoning model group was fed a normal diet mixed with 140 mg/kg CdCl2;the blank group was fed a normal diet;the Ganoderma triterpenoids treatment group was fed a normal diet containing 140 mg/kg CdCl2,while each was given 0.5 mL of Ganoderma triterpenoids solution?20 mg/mL?;Ganoderma triterpenoids group was fed with normal feed,and each was given 0.5 mL of Ganoderma triterpenoids solution?20mg/mL?.The chickens were euthanized on the 20th,40th,and 60th day respectively,and the testes and kidneys were removed.Determination of testis and kidney organ coefficient,detection of cadmium in testis and kidneys,detection of antioxidant enzymes?SOD,GSH-Px?and malondialdehyde?MDA?content using biochemical kits,real time The expression of cytokines?TNF-?,IL-1?and IL-6?and heat shock protein?HSP 60,HSP 70 and HSP 90?mRNA were detected by fluorescence quantitative assay.The heat shock protein?HSP 60?was detected by Western blot.Protein expression of HSP 70 and HSP 90).The results showed that in the cadmium poisoning model group,the viscera coefficient of the testes was significantly lower?P<0.05?,the viscera coefficient of the kidneys was significantly higher?P<0.05?,and the cadmium accumulation in the testes and kidneys was significantly increased compared with the blank group.?P<0.05?;Antioxidant enzymes?SOD,GSH-Px?activity in the testis and kidneys was significantly decreased?P<0.05?,malondialdehyde?MDA?content was increased?P<0.05?;inflammatory cytokines in testis and kidneys(mRNA expression of TNF-?,IL-1?,and IL-6 was significantly increased?P<0.05?;mRNA and protein expression of heat shock proteins?HSP 60,HSP 70,and HSP 90?in testis and kidneys were significantly increased?P<0.05?.Compared with the cadmium poisoning model group,the Ganoderma triterpenoids treatment group significantly increased the testicular coefficient?P<0.05?and the kidney coefficient significantly?P<0.05?;the cadmium accumulation in the testis and kidneys decreased significantly?P<0.05?;Antioxidant enzymes?SOD,GSH-Px?activity was significantly increased?P<0.05?,Malondialdehyde?MDA?content was significantly decreased?P<0.05?;Chicken testis and kidney inflammatory cytokines?The expression levels of TNF-?,IL-1?,and IL-6?mRNAs were significantly decreased?P<0.05?;mRNA and protein expressions of heat shock proteins?HSP 60,HSP 70,and HSP 90?in the testes and kidneys were also significantly decreased?P<0.05?.There were no significant differences in the testis and kidney parameters between the Ganoderma triterpenoids Sancha chickens and the blank chicks.The results of this experiment show that Ganoderma triterpenoids has significant protective effect on the toxic damage of testis and kidney caused by cadmium,and the experimental results provide a theoretical basis for further prevention and treatment of cadmium poisoning.
