Therapeutic Effect Of Sophora Flavescens Antidiarrheal Capsule On Canine Parvovirus Disease

resistance,it has become a new topic to develop new,effective,non-toxic and no side effects of Chinese herbal medicine against canine parvovirus.Sophora flavescens antidiarrheal Capsule is composed of capsule,Radix Paeoniae Alba.Radix Sophora etc.It has heat-clearing and damp-drying drug,cooling blood and stop diarrhea effect.The purpose of this study is to investigate the therapeutic effect of Sophora flavescens antidiarrheal Capsule on canine parvovirus disease by cell test,animal test and clinical medication.Text 1 Determination of the content of active components of Sophora flavescens antidiarrheal capsule by HPLCRP-HPLC method was developed for the determination of matrine,Oxymatrine,Dehydrocostus lactone and Costunolide.Used Phenemonex C18(250 mm×44.6 mm,5?m)Chromatographic column,The mobile phase of matrine and Oxymatrine matrine with lOmmol/L aqueous solution of acetic acid(pH8)-methanol gradient elution,Detected at 210 nm.Flow rate:1mL/min;The mobile phase of paeoniflorin with acetonitrile-0.1%phosphoric acid solution(14:86),Detected at 230 nm,Flow rate:1mL/min;The mobile phase of methanol and water(65:35)for the removal of Dehydroepiandrosterone and costunolide,Detected at 225 nm,Flow rate:0.65mL/min.Results matrine was 2.2mg/g,Oxymatrine as 1.05mg/g,paeoniflorin as 9.1mg/g and 13.5mg/g,and the content was too low.Text 2 Study on the effect of Sophora flavescens antidiarrheal Capsule on canine parvovirusIn order to observe the effect of matrine flavescens antidiarrheal capsule in vitro on canine parvovirus,using FK81 cell as the carrier,through the indirect immunofluorescence assay(IFA),into cells of canine parvovirus using fluorescein labeled and observed under fluorescence microscope,fluorescence analysis with ImageJ software.The results showed that the infection rate of canine parvovirus in FK81 cells could be significantly reduced by adding the traditional Chinese medicine to the virus group and the traditional Chinese medicine and virus infection group,The average reduction of adding Chinese medicine then virus group was 36%,and the average reduction of the infection group was about 46%;There was no significant difference in the expression of FK81 in the infected group and adding virus then Chinese medicine virus group group.The results showed that the solution of Sophora flavescens antidiarrheal capsule can effectively prevent the adsorption and invasion of canine parvovirus,and could not inhibit the replication of canine parvovirus.Text 3 Clinical observation on the treatment of canine parvovirus with Chinese and Western MedicineIn order to observe the effects of matrine flavescens antidiarrheal capsule in the treatment of canine parvovirus disease,with 7 dogs from horizontal infection as an animal model,divided into Chinese medicine group,Western medicine group,blank group and positive group were 3,3,1and 1,matrine flavescens antidiarrheal capsule and Western medicine with dysentery the treatment,clinical symptoms,pathological anatomy and histopathological examination,electron microscope.Results:Chinese medicine and Western medicine can control the canine parvovirus disease,including Chinese medicine group showed no blood dysentery symptoms,histopathological observation of intestinal tissue and no obvious bleeding,and maintain the integrity of the intestinal villus structure,showed that matrine flavescens antidiarrheal capsule can inhibit dysentery caused by canine parvovirus the intestinal mucosal injury and hemorrhagic lesions,so as to achieve the healing of canine parvovirus disease.Text 4 Clinical trial on the treatment of canine parvovirus disease with Sophora flavescens antidiarrheal CapsuleIn order to further investigate the effect of matrine flavescens antidiarrheal capsule in treating canine parvovirus disease.Methods:used the cases of canine parvovirus disease were treated by animal hospital,The contrast test was made between Western medicine and Combination therapy of Chinese medicine and Western Medicine.Statistics the cure rate,cure course and so on of two kinds of medicine.Results:32 cases of canine parvovirus disease treated with western medicine,The cure rate was 75%;32 cases of canine parvovirus disease treated with combination therapy of Chinese medicine and Western Medicine,the cure rate was 87.5%;and the average cure treatment treated with combination therapy of Chinese medicine and Western Medicine treatment for 5.32d,lower than the average cured 6.29d pure Western medicine,combination therapy of Chinese medicine and Western Medicine treatment group of dogs to cure the average number of days of blood stasis was 3.86d,significantly lower than the western medicine group 5.38d(P<0.05).Conclusion:matrine flavescens antidiarrheal capsule has good therapeutic effect on canine parvovirus disease.
