Cucumber is a most important vegetable in the world.However,cucumber production was restricted due to long-term unscientific cultivation method,such as monoculture system and the abuse of chemical fertilizer.Fusarium wilt of cucumber is one of the most destructive worldwide disease of cucumber.The Fusarium wilt was caused by increased pathogenic microflora inhibited the growing of beneficial predominant microflore in the soil.There are many ways to prevention and control of Fusarium wilt.Among them,biological control is considered the most promising method.Some studies have shown that organic fertilizer contains active substances and nutrient elements.That can improve crop root soil microbial activity.In this paper,we carried out various exploration about control blight of cucumber and drew the results as follows:(1)The pot experiment to study the influence of different concentrations of bio-organic fertilizer(BIO)on the cucumber yield,cucumber Fusarium wilt and soil microflora alterations.The results show that compared with chemical fertilizer controls(CK),use of high concentrations of BIO can significantly decrease incidence of cucumber fusarium wilt,increase production and soil fertility.The cucumber yield of BIO 10 and BIO20 is about 5 times of CK,BIO2.5 and BI05.And the real-time PCR results show that the relative number of F.oxysporum f.sp.cucumerinum of the soil decreased with the increase of BIO concentration.And the relative number of B.subtilis SQR9 of the soil increased with the increase of BIO concentration.(2)Biolog analysis showed that compared with CK,use of high concentrations of the BIO could increase the activity of soil microorganisms and the carbon source utilization ratio.However,the use of low concentration of BIO could reduce its.(3)High-throughput sequencing analysis results show that compared with CK,use of high concentration of BIO could significantly increase the number of Kaistobacter,Hyphomicrobiu and Aspergillus in the soil.At the same time,use of high concentration of BIO could decrease the number of Cladosporium,Monosporascus and Phoma in the soil.(4)Experimental results in small plots showed that compared with no fertilizer control and treatments with manure,the yield of applying BIO treatments increased by 23.11%and 16.31%,respectively.The disease incidence of applying BIO treatments were lower than 10%.Use of BIO could significantly increase the soil fertility.(5)Rhizosphere soil microorganism determination results show that compared with no fertilizer control and other treatments,the processing of applying BIO increased the total number of soil microorganisms.At the same time,use of BIO could increase the number of Pseudomonas and Rhizobia in the soil.
