The dried and ripe seeds which picked from Aesculus wilsonii Rehd.have the effect on soothing liver and regulating Qi flow,protecting the stomach and relieve the pain.Modern studies have shown that Aesculi Semen has therapeutic effect on treatment of cerebral hemorrhage,cerebral edema,limb swelling,burns,peripheral facial neuritis,meniere's syndrome the same as cancer.In recent years,with people's intensive studying of its curative efficacy,market demand is increasing.At present,the quality of Aesculi Semen is unstable and has bad effect on drug safety,because of the reducement of wild resources and market supply.So,this study analyses the ecological suitability of the basal plants of Aesculus wilsonii Rehd.on the basis of the Global Geographic Information System for Medicinal Plant and the absorption of nutrient elements in different growth stages,and research the effects of different storage methods and different size on seeds viability,then,to develop system fertilizer program of a different growth period,last,using the main chemical composition analysis method to study the optimum growth period and guide the cultivation of traditional Chinese medicine.Global geographic information system for medicinal plant was used to study the global ecological suitability of the Aesculus wilsonii Rehd.The results showed that the optimum area of Aesculus wilsonii Rehd.growth mainly involved in China,North Korea,the United States,Brazil,Australia,France,Italy and Portugal,of which the ecological suitability regions in China,including Hubei,Anhui,Jiangsu,Hunan,Sichuan,Yunan,Guzhou,Shanxi,Henan,Guangxi,Fujiavn province,etc.In this experiment,the effects on different storage methods and different size of seeds viability of Aesculus wilsonii Rehd.by using TTC method was studied.The results showed that the optimal determination conditions for the viability of Aesculus wilsonii Rehd.was staining for 11 h to 13 h with0.2%TTC solution.During the seeds breeding process,the best condition for storage of Aesculus wilsonii Rehd.seeds was storage-in-sand with large particle size seeds.In research,we analysis on nitrogen,phosphorus,potassium and zinc uptake and distribution in different growth stages of Aesculus wilsonii Rehd.we found that the requirement of nitrogen was the highest in the plant growth period and minimum requirement was phosphorus;The requirement of fertilizer increased during plants matured;Compared to the element content in soil with plant,we found that phosphorus and zinc in the soil were low,so it should be supplement during normal fertilization.In this paper,the content of Aescin A and escinate B were analyzed by HPLC with different growth years and different growth stages.The specific chromatographic conditions are as follows: the separation was achieved on Themo BDS HYPERSIL-C18 column at 30?,using methyl cyanide-0.2%phosphoric acid solution(37:63,v/v)as the mobile phase at a flow rate of1.0mL/min.The detection wavelength was 250 nm,and the sample volume was 10?L.Based on comprehensive evaluation,Aesculi Semen growth after12 years in white dew good harvest.
