Study On The Quality Control And Pharmacodynamics For Compound Herba Houttuyniae Granule

Compound herba houttuyniae granule was made according to the basic theory of treatment based on syndrome differentiation in traditional Chinese veterinary medicine and the major functions of Chinese medicine.The antibacterial tests in vitro and acute toxicity test have been done for determining the security and antibacterial effects of its compound herba houttuyniae granule in order to provide reference for its clinical application.1.Antibacterial activity in vitro of single medicine and compound prescriptionUsing the classical double tube dilution method determination the MIC and MBC of herba houttuyniae,radix astragali,fructus forsythiae,flos lonicerae japonicae and compound herba houttuyniae against Staphylococcus aureus,Streptococcus agalactiae,Escherichia coli,Salmonella and Pasteurella multocida.The results showed that the effect of compound herba houttuyniae was greater than the single medicine.2.Preparation and quality control test of compound herba houttuyniae granuleThe herba houttuyniae,radix astragali,fructus forsythiae and flos lonicerae japonicae are 42 percent,9 percent,4.5 percent and 4.5 percent,and beta-cyclodextrin 40 percent,which is prepared by hand wet legal system for compound herba houttuyniae granule.The granule are between 200 ?m~2000 ?m,and the color is tan.The compound herba houttuyniae granule of size,loss on drying,melt index and other indicators were investigated,determined the content of total flavonoids and polysaccharides in compound herba houttuyniae granule by UV.The results showed that the compound herba houttuyniae granule of size,loss on drying,melt index and other indicators were in line with the Veterinary Pharmacopoeia;the average content of total flavonoids was 39.1 mg/g,and the average content of polysaccharide was 281.9 mg/g,which was 97.8 % and 94.0% respectively,meet the requirements of pharmaceutical preparation.3.In vitro antibacterial test of the compound herba houttuyniae granuleUsing the classical double tube dilution method determination the MIC and MBC of compound herba houttuyniae granule against Staphylococcus aureus,Streptococcus agalactiae,Escherichia coli,Salmonella and Pasteurella multocida,the results showed that MIC(mg/mL)was 45.71,45.71,182.86,365.71 and 182.86 respectively;MBC(mg/mL)was 45.71,91.43,365.71,731.43,365.71.The results of the experimental results showed that the compound was not affected by antibacterial effect.4.Acute toxicity test of the compound herba houttuyniae granuleThe acute toxicity test was did to evaluate the safety of compound herba houttuyniae granule,and the results showed that the LD50 is greater than 10000 mg/kg.According to the stipulations of the WHO,if the LD50 is 5000~15000 mg/kg,the compound herba houttuyniae granule is the actual non-toxic drug.The security of the compound herba houttuyniae granule is good,it belongs to actually non-toxic medicine,which can be safely used in clinical treatment.
