Plants non-specific lipid transfer proteins richly express in plants,which involved in many biological activitis,but few was known in wheat.In this study,we cloned and analyzed three TaLTPI genes and promoters from wheat,and the main work is as follows:(1)Three TaLTPI gene were cloned from wheat and overexpressed in Arabidopsis resulted in longer root length compared with wild type plants under chilling condition.The three TaLTPI genes,TaLTPIb.1,TaLTPIb.5,and TaLTPId.1,all constitutively enhanced the tolerance to chilling stress.(2)Fused with EGFP,all the three TaLTPI proteins were shown to localize on the plasma membrane in transgenic tobacco,although a signal in chloroplasts was also observed for TaLTPId.1.TaLTPIb.1,TaLTPIb.5 were up-regulated by drought,chilling and wounding,but depressed by darkness.However,TaLTPId.1 showed a relatively distinct expression profile compared with TaLTPIb.1 and TaLTPIb.5,which was up-regulated by MeJA,darkness,implying different biological roles.(3)The genomic distribution showed that TaLTPIb.1 and TaLTPIb.5 were located on chromosomes,3B and 3D,while TaLTPId.1 was observed to be located on chromosome 5D.(4)We tested the promoter activity of TaLTPIb.1 in a transgenic brachypodium,which was observed mainly in young leaves,shoots,and spikes,but was not observed in roots.(5)The activity area of the promoter were located between upstream of-400 bp and-256 bp incuding three important cis-acting element,GC rich motif,CA rich motif and CATC motif,and GC rich motif was the most important element in epidermis specific expression pattern.
