The growth of plants is affected by drought, frost, salinization or acid of the soil, and inadequate environmental stress.They restrict the output of field crops, and the development of agriculture. But in all of these stress, the lack of moisture content most widely exists. Plasma membrane and thylakoids are damaged during freezing,because the plasma membrane. As it has been found that, in general, plant survival during freezing is limited by the stability of cellular membranes. So the multi-resistant gene from the plant is very important for resisting inadequate environmental stress.Chimonanthus praecox (L.) Link(Calycanthaceae) is a deciduous shrub , an endemic plant lived in China. It is famous for its landscape and medicinal value. Chimonanthus praecox (L.) Link could bloom at low temperature. So we believed that there must be some kinds of antifreeze materials in wintersweet.It is shown in RT-PCR that,the nsLTPs expression of the wintersweet after a freeze–thaw cycle for five days is much richer than the nsLTPs expression of the wintersweet ,which always in 20℃. We summarize our knowledge of those cold-regulated proteins that are ableto stabilize membranes during a freeze–thaw cycle in the wintersweet. And nsLTPs are thought to be involved in the transfer of lipids through the extracellular matrix for the formation of cuticular wax. The cuticular wax in wintersweet is more high than tobacco that is considered as a drought-tolerant species. And the rate of actual weight loss due to epidermal conductance from leaves excised from the wintersweet is lower than the tobacco. we provide strong correlative evidence that nsLTPs plays an important role in inadequate environmental stressI had designed the primer of the nsLTPs and cloned three nsLTPs genes.After that I got the open reading frame (ORF) sequences . We find that a high content of hydrophobic amino acids for bioinformatics analysis and the nsLTPs are characterized by their small molecular mass and eight conserved Cys residues that form four disulphide bridges in a highly conserved pattern. The nsLTPs including cryoprotectin, are also characterized by their high stability . we have constructed plant expression vector pTEV7-CpnsLTP-1for the study of biological function, We transformated Arabidopsis thaliana with Agrobacterium and on MS medium containing hygromycin sieve transgenic from T0 generation seeds,and finally I got the transgenic plants.It could be an important theoretical significance and economic value to know more functions of the nsLTPs .
