Streptococcus suis is zoonosis which brought big economic losses on the global swine industry and the health of the public. Streptococcus suis have 33 serotypes (1-34, 1/2 serotype). In order to rapid identification of streptococcus suis serotype, the Multiple PCR diagnostic method was established. The main results were as follows:(1)Using bacterial culture and PCR identification,417 strains of streptococcus suis were obtained from 2013 to 2015, which included 26 species of serotypes (such as 1 or 14?2 or 1/2?3?4?5?6?7?8?9?10?12?13?14?15?16?19?20?21?23?25?26?28? 29?31?33).The detection rate of serotype 2 (1/2)?1 (14)?3?7?8?9?10?21 and 23 was 12.7%?8.63%?8.87%?6.0%?6.95%?5.0%?4.8%?4.56%?4.56% respectively, and other serotypes all less than 4%.(2)By Specific primers and matched group which included mixed serum, Staphylococcus aureus, Escherichia coli et al. The multiplex PCR serotyping assay for streptococcus suis which included three-Serotype PCRs-GDH,2,3 and 1,7,9 and one Four-Serotype PCR-8? 10?21?23. The result showed that specific and sensitive of three kinds of multiplex PCR was very well.(3)263 samples of the nasal swabs and 150 samples of tonsil were detected by three kinds of multiplex PCR. The result showed that:relevance ratio of samples nasal swabs and tonsil was 67.7% and 75.6% respectively for GDH; 10.0% and 11.2% for 1 serotypes; 25.5% and 25.0% for 12 serotypes; 11.0% and 15.6% for 3 serotypes; 11.4% and 11.9% for 7serotypes;8.7% and 11.2% for8 serotypes;3.9% and 7.5% for 9 serotypes;4.2% and 5.6% for 10 serotypes.The result showed that quadruple PCR-identification was highly specific and sensitive, could be applied on rapid diagnosis of Streptococcus suis.
