Efficacy Of Compound Chinese Herb Exacts Preparation On Anti-newcastle Disease Virus And Quality Control

Pigeon Newcastle Disease(ND),also known as Pigeon Plague or Pigeon Paramyxovirus type 1(PPMV-1),is an acute,febrile,highly contagious septic disease caused by Newcastle Disease Virus(NDV).For the prevention and treatment of the disease,extensive use of antibiotics and chemical drugs leads to the emergence of drug resistance and drug residues.Moreover,with putting forward of ecological farming and green food,the regulation has been drawn in China,for feed and food,the use of veterinary clinical antibiotics and chemical drugs is strictly prohibited.It is generally known that traditional Chinese medicine has the advantages of antiviral effect,little side-effect and no resistance.Not only could it improve immunity of the organism,but also it can promote growth and improve performance.In this study,under the guidance of Chinese veterinary medicine theory,10 kinds of Chinese herb exacts such as flos lonicerae,radix scutellariae,fructus forsythiae,hypericum perforatum etc were used in compound Chinese herb extract preparation,to study the ability of antiviral,immunological enhancement and quality detection standard of compound Chinese herb extract preparation.The results are as follows:1.The hemagglutination titer in prevention and treatmentgroup with high doses(including crude drug 2 g/mL)and middle doses(including crude drug 1 g/mL)of compound Chinese herb extract preparation was significantly reduce(P<0.01),in which the effect of middle dose group(including crude drug 1 g/mL)compound Chinese herb extract preparation is remarkable.2.Compound Chinese herb extract preparation could improve the antibody titer of NDV vaccine.The antibody titer of the middle dose group(including crude drug 1 g/mL)of compound Chinese herb extract preparation was notably different(P<0.05),and the bursa of organ index in the middle dose group was notably different(P<0.05).Lymphocytes lined up tightly in the immune organs,and the number of lymphocytes was higher than that of the positive control group.3.Baicalin was test components to study the quality detection of compound Chinese herb extract preparation by TLC and HPLC.TLC results showed that baicalin for the test and reference substance in the corresponding position appeared in the same color spots,negative reference substance without interference phenomenon;HPLC results show that baicalin hasgood linear relationship in the concentration of 10 ~ 200 ?g/m L,and the content of baicalin was 3727.41 mg/g.The retention time of baicalin peak was consistent with that of the control,and the negative reference substance without interference phenomenon.Compound Chinese herb extract preparation has a good inhibitory effect on Newcastle disease virus,which can significantly improve the NDV vaccine antibody titer and bursa of organ index,and make the immune organ lymphocytes arranged closely and the number increased.The method of TLC and HPLC detection is specific and can be used to detect the contents of baicalin in compound Chinese herb extract preparation.
