The complex polysaccharide has many biological activities and is the best immunomodulator with high quality and non-toxicity,small side effects and wide source.At present,the trend of drug research is due to the development of chemical synthetic drugs in the direction of natural drugs,and research on polysaccharides of traditional Chinese medicine has begun to receive attention.Studies have shown that the complex polysaccharides are more effective than the polysaccharides alone in the magnitude of immune activation or on the level of immunoregulation due to the synergistic effects of polysaccharides.Therefore,in recent years,research on polysaccharides has been progressing toward the research of pharmacological activities after combination of various polysaccharides.According to the pharmacological action,the pharmacological activity combines the individual polysaccharide components extracted from different plants in a certain ratio,thereby preparing a complex polysaccharide having synergistic effects between the polysaccharides.In this experiment,four traditional Chinese medicines containing polysaccharides were selected as the research object of drug efficacy tracking.Four kinds of Chinese medicine polysaccharides were extracted by water extraction and alcohol precipitation method,and the four polysaccharides were purified,and the effective polysaccharide combinations were mixed in a certain proportion to make a composite polysaccharide.To observe the antiviral activity of four polysaccharides and complex polysaccharides in vitro,to detect the antiviral effect of the individual and complex polysaccharides in vitro,and to find the best active polysaccharides with anti-chicken Newcastle disease virus and their dosage.The test results are as follows:1.Fomes Officinalis Ames Polysaccharide(FAP),Lagenaria siceraria polysaccharide(LSP),Ferula sinkiangensis polysaccharide(FSP),and Artemisia rupestris polysaccharide(ARP)were extractedfrom four different traditional Chinese medicines by water extraction and alcohol precipitation method,and determining the content.The content of these polysaccharides and the yield of the polysaccharides were 45.3%,28.6%,43.8%,39.9%,and 2.6%,7%,4.9%,and 10.6%,respectively.2.In order to study the anti-Newcastle disease virus activity of four traditional Chinese medicine active ingredients in vitro,chicken embryo fibroblasts were cultured in vitro,and the antiviral activity of polysaccharides was determined by MTT assay.The results showed that all four polysaccharides had certain antiviral activity,among which LSP And ARP has a better blocking effect on NDV,and the virus inhibition rate can reach 33.81%and 29.66%.The four polysaccharides such as FSP,LSP,ARP and FAP have excellent inhibitory effect on NDV,and the virus inhibition rate can reach 75.28%.63.59%,63.25%,44.95%;FAP and LSP polysaccharides have a direct killing effect on NDV,and their virus inhibition rates can reach 24.07%and 14.51%.3.In order to obtain the pure products of the four traditional Chinese medicine polysaccharides,the four crude polysaccharides were decolorized and purified.The purification and yield of the polysaccharides showed that:①The decolorization effect of the macroporous resin D900 showed that the content of the four polysaccharides before and after purification The yields are(FAP,LSP,FSP,ARP),respectively(the same below)54.6%,30.9%,57.7%,44.01%and 2.6%,7%,4.9%,10.6%.② The results of four kinds of traditional Chinese medicine polysaccharides were degreased with petroleum ether,ethanol,and the protein was removed by Sevage.The results showed that the polysaccharides of FAP,LSP,FSP and ARP were improved after purification,and the groups were identified by infrared spectroscopy.The polysaccharides all have characteristic peaks of polysaccharides.Studies have shown that after purification of the four polysaccharides,there is a certain increase in the polysaccharide content of each group and avoiding the influence of the test results due to the large number of impurities.4.In order to study whether the polysaccharide combination can enhance the polysaccharide activity,the four polysaccharides were mixed into a composite polysaccharide to observe the antiviral activity of the polysaccharide in vitro.The results showed that the compound polysaccharides had antiviral effects and had certain therapeutic and preventive effects on NDV.Among them,the anti-NDV activity was the strongest when the virus was first added and the polysaccharide was added first,especially compound Ⅷ,complex Ⅰ,complex Ⅱ,Polysaccharides such as complex Ⅲ,complex Ⅸ,complex Ⅳ,and complex Ⅴ have the best anti-NDV activity,so these complex polysaccharides are polysaccharides with excellent antiviral effect.It is again demonstrated that the polysaccharides can be mixed to improve the anti-NDV activity of polysaccharides.
