Papaya(Carica papaya Linnaeus)is one of the most important fruit trees in many tropical and subtropical countries.It is very difficult for large-scale production of papaya with conventional ways of papaya seedling.The lateral buds of fruit-type papaya were taken as explants from the field preferred.These explants were incubated on MS medium,supplemented with various concentrations of MS macroelement,auxin,cytokinin and additive.The sterile shoots differentiation and propagation,root differentiation were studied.The best transplanting substrate were obtaining by screening test.The system of in vireo rapid propagation of fruit-type papaya was established preliminary.Based on this system,pre-production for "Zhong bai" fruit-type papaya was carried out.The key is to study the effect of season,ways of multiplication culture,rooting culture vessel and smelting plants of rooting seedling on pre-production.Production technologies was optimized and best combination was processed in order toachieve commercial production technical level.In this study,the best season to derive lateral buds was in autumn,the best induction results were obtained by using the crown and the following 1 to 2 sleeping bud,the best disinfection procedure of explants was:with 75%alclhol for 20 seconds,and then soaking with 0.1%mercuric chloride in 12mimutes,finally rense for 5 to 6 times with sterile water.The disinfection process induced the success rate of 56.67%.The original medium for best sterile shoots was:MS+6BA0.4mg/L+NAA0.1mg/L.The best strenghening bud medium was:3/2MS+6BA0.2mg/L+NAA0.1 mg/L+Su30g/L,multiplication coefficient was 3.22.the effective buds coefficient of 2.18.Root induction culture medium by the method of two-step rooting effect was the best,rooting rate was 95%,and the percent of pass was up to 92.22%,the first step of medium(MS+IBA2.0mg/L)induced 5 to 7 days,and then go to step two hormone-free medium(add 1g/L activated carbon)to induce rooting.Transplanting the matrix to light matrix effect was best,the survival rate was 90%,the percentage of seedling rate was 80%.With the materials and technology obtained above.The pre-production of "Zhong bai"fruit-type papaya produced more than 180000 strains rooting seedling.All the indexes meet the requirements of large-scale production.Multiplication coefficient was 2.88?3.91,of which the effective buds coefficient was 1.72?2.32.Rooting rate 83.89%?93.89%,the percent of pass was 81.67%?93.33%,direct cost was ? 0.4317 per plant root container seedlings production.Transplanting survival rate 71.40%?92.53%,the rate was 68.00%?90.07%,direct production cost was ? 0.4267 per plant transplanting seedling production.Through the pre-production showed that the best season were summer and autumn because of their production output and best quality.Transgenerational seedling production of temperature control was the most economic cultivation mode in 15?30?.Light was scattered light was given priority to with a natural,gradually transition to the week light from light,had a little effects on strenghening bud culture,but greatly save energy consumption,make full use of the training space.Rooting seedling production can need not open smelting plants,the transplanting survival rate,can simplify steps.
