Objective:This study aims to extract the beneficial microorganism from pigsty environment-Bacillus subtilis, domesticated, cultivated, and identified, to explore its antibacterial properties. Finally developed a micro ecological preparation of Bacillus subtilis that mainly applied to environmental improvement, reduce the incidence of the disease.Test method:1.Collect natural feeding pigs'fresh feces and urine, isolating and culturing its bacteria, then identified by the microscopic and molecular biology technology,16SrDNA sequencing to determine required bacillus subtilis.2.Simulating piggery environment, culturing the identified Bacillus subtilis, finally get a kind of bacillus subtilis which can grow fast in harsh environment pigsty.3.Doing vitro antibacterial test with Escherichia coli,Salmonella. Staphylococcus aureus by domesticated bacillus subtilis.4.Mix the bacillus subtilis and montmorillonite by certain proportion, put the mixture into the pigsty, confirm its effect.Results:1.The bacillus subtilis screened from the pigsty have the 99% homology with bacillus subtilis in NCBI on 16SrDNA gene sequence.2.Contrasting the domesticated bacillus subtilis from pigsty and normal bacillus subtilis:the domesticated bacillus subtilis grows fast in 12h. and the normal bacillus subtilis grows slow. The domesticated bacillus subtilis have the 99% homology with bacillus subtilis in NCBI on 16SrDNA gene sequence.3.The vitro antibacterial test shows that the domesticated bacillus subtilis have a good effect on restrain Escherichia coli. Salmonella and Staphylococcus aureus. antibacterial time can reach more than 48 hours.4.Put the mixture of domesticated bacillus subtilis and montmorillonite into pigsty, the number of bacteria in the air and on the ground are significantly reduced.Conclusion:The domesticated bacillus subtilis from pigsty can grow fast in harsh environment, and have a good effect on restrain bacterial, in order to improve the environmental protection effect, herd health.
