In order to verify the immunoenhancement effect and the security of Huozhen powderfor, so as to provide test basis for its clinical application, Huozhen powder's pharmacodynamics and security tests were systematically conducted.1. Pharmacodynamics testNon-specific immunoenhancement test:40 mice were randomly divided into 4 groups: group I-III were the trial groups, fed with the feed added with 2425mg/kg,4850mg/kg and 9700mg/kg Huozhen powder, while group IV was the control group, fed with the normal feed.7d later, the daily gains, feed efficiencies, exhausted swimming times, spleen index and thymus index of the mice in each group were determined. Results showed,, the growth performance of mice in each trial group had no significant difference compared with that in the control group, while the spleen index and the thymus index increased, the exhausted swimming times of mice in group ? and ?increased, and among them, spleen index and the thymus index in group II increased more than those in the control group.Specific immunoenhancement test:200 mice were randomly divided into 5 large groups, named dose groups of BSA 0.1 ?g,0.3 ?g and 0.9?g,2.7 ?g and 2.7 ?g per mouse; and each large group were randomly divided into 4 small groups, small group 1-3 were the trial groups, fed with the feed added with 2425mg/kg,4850mg/kg and 9700mg/kg Huozhen powder, and small group 4 was the control group, fed with the normal feed. All the above mice were fed for 14 days continuously, BSA were inoculated on 7th day, and blood samples were collected on 14th day to determine the titer of anti-BSA antibody in serum. Results showed that, the best vaccination dose of BSA on mice was 0.3?g/mouse, and the titer of anti-BSA antibody under this dose of BSA could reach 0.277±0.059. Among all small groups, the titer of anti-BSA antibody in group 1 was the highest, reaching 0.300±0.059, and significantly higher than that in the control group.It promped that, Huozhen powder had a better enhancement effect on non-specific and specific immunity in mice.2. Acute toxicity testPre-tcst:20 mice were randomly divided into 5 groups, group 1?4 were the trial groups, fed the feed respectively added with 5000,1000,200,40 mg/kg Huozhen powder for 1 day; and group V was the control group, fed with normal feed. All mice were observed for continuous 14 days. If all the mice in the test didn't die or appear toxic reaction, the formal test would be conducted.Formal test:50 mice were randomly divided into 5 groups, group I ? IV were the trial groups, garaged with 0.4mL saturated solution of Huozhen powder and its 1:2,1:4, 1.'8 diluents per time,1 time/2h,3 continuous times; while group V was the control group, garaged with 0.4mL saline solution by the same method. Toxic reaction and death situation were observed and recorded.Results showed that, all mice in the pre-test and the formal test didn't die, nor toxic reactions occurred.It prompted that, LD50 of Huozhen powder was above 5000 mg/kg, and it was actually non-toxic.3. Sub-chronic toxicity test400 mice and 160 rats were randomly divided into 4 groups respectively, group I-III were the trial groups, fed with the feed added with 1.2 mL/g bw(mouse) and 0.02 mL/g bw(rat) saturated solution of Huozhen powder and its 1:2,1:4,1:8 diluents; while group V was the control group, fed with the feed added with the same volume of saline solution. The behaviors and characteristics of all mice and rats would be observed and recorded, as well as toxic reaction and death situation. On the 45th day, livers, kidneys, hearts, spleens, lungs and other organs of all mice and rats were collected to observe the pathological changes and slice to observe the histopathologic changes; the blood ALT, AST, UA and CREA were determined. The results showed that, all mice and rats in the test didn't die and no toxic phenomenon occurred; all the blood biochemical indexes were in normal range; all organs had no visible and histological pathological changes. It prompted that, Huozhen powder han no sub-chronic toxicity on mice and rats.4. security test on target animal-pig5021-day-old hybrid (LandraceŚLarge Yorkshire) piglets were randomly divided into 5 groups, group ??? were the trial groups, fed with the feed added with 1000 g/t,3000 g/t and 5000 g/t Huozhen powder respectively, group IV and V were the control groups, fed with the feed added with 1000 g/t Astragalus polysaccharide and normal feed respectively. All the piglet were fed for 14 continuous days, and their signs, behaviors, dungs and urines were observed and recorded. On the 14th day, blood samples were collected to determine the blood cell count, ALT, AST, UA and CREA.1 piglet from each group was killed to observe organs pathological changes, and its liver, heart, spleen, lung, kidney and other organs were taken to slice and observe the histopathological changes. The results showed that:the behaviors, signs dungs and urines of every piglet from every group were normal, the blood test index as well as the blood biochemical index of the trial groups had no significant difference from those of the control groups, and every organ from the trial groups had no visible and histopathological changes. It prompted that, Huozhen powder was safe on target animal-pig.To sum up, Huozhen powder could enhance the specific and non-specific immune function of mice, had no acute and sub-chronic toxicity on mice and rats, and was safe on piglets.
