| Clerodendrum trichotomum Thuhb, a deciduous shrub or small tree, belongs to genus Clerodendrum, family Verbenaceae which is widely distributed from north China, east China, central south to southwest provinces in China, as well as distributed in Japan, Korea and the Philippines. C. trichotomum is a rare woody wild vegetable with unique flavor, which could be used in greening, landscaping, aromatization, medicine and food. It has great utilization value and good marketing potential. In recent years, there is no seed can be collected because of intemperance in pluck of buds, which leads to serious natural resources destruction. In order to meet the demand for the resource by public and ensure sustainable utilization of C. trichotomum, it is necessary to propagate artificially. Compared with traditional propagation methods, tissue culture can produce a lot of plants in a short time, meanwhile, this method ensure good offspring traits and the same biological characters between individual.In this study, germplasm resources of C. trichotomum in Yunnan were investigated and collected, which provide materials for further developing and utilization. Comparative study of explant types, disinfecting methods, culture media, effects of different growth regulators on bud inducing and rooting and effects of different planting media on transplanting were conducted.The experiment and results are as follows:1. Survey on permplasm resources of C. trichotomum in YunnanDuring 1 year’s investigation and collection, distribution, cultivation and marketing of C. trichotomum in Yunnan have been investigated. Only one variety- C. Trichotomum with smooth leaves was found after surveying. 60 germplasm resources were collected and grown in nursery of Yunnan Agricultural University.2. Establishment of tissue culture in vitro of C. trichotomum(1) Compared with different explants(leaf, stem tip and stem segment), the result showed stem segment was the optimal explant. Survival rate and inducing rate of stem section are much higher than that of leaf and stem tip.(2) Effect of sterilization showed that the best result obtained from treatment with 75% alcohol for 30 s followed with 0.1 mg/L mercuric chloride for 7 minutes.(3) Media for bud start of C. trichotomum were compared by microcutting. The optimal inducing medium was MS + 0.2 mg/L 6-BA + 0.1 mg/L IAA.(4) MS, WPM and B5 media were compared for shoot proliferation and growing. For shoot proliferation, WPM > B5 > MS. The best medium was WPM + NAA 0.1 mg/L + 6-BA 1.0 mg/L.For shoot growing, MS medium is best.(5) Rooting media were tested. All growth regulators could significantly improve rooting rate of C. trichotomum, though different rooting time and root growth situation along with different growth regulators were found. The optimal medium for rooting was MS+1.0mg/L IBA, which produced the best results: rooting early, maximum quantity of roots, the highest rooting rate of 85%, and shoots grown well.(6) Test of rooting substance. As far as survival rate, vermiculite: peat = 1:1 was better than perlite: vermiculite = 1:1and perlite: peat = 1:1. |
