The aim of this study was to investigate the effects of traditional Chinese medicine formula(TCMF) and some single medicines on the genetic expression of IFITM3 in 10-day-old broilers, the effects of TCMF with different preparation and method of administration on the genetic expression of IFITM3 in broilers and the effects of TCMF with different doses on the genetic expression of IFITM3, the contents of IFN-?, IFN-? and IFN-? in serum, the detoxification amounts and the blood biochemical in NDV broilers in order to research the relationship between IFN and IFITM3 further. In order to provide theoretical basis for developing antiviral TCMF by seeking the mechanism of TCMF on NDV.There were 3 parts in this study. Part 1: to study the effects of TCMF and single medicine on the genetic expression of IFITM3 in 7 trial days. 10-day-old 210 broilers were randomly divided to 7 groups, including TCMF group, folium isatidis group, honeysuckle group, fructus forsythia group, Scutellaria baicalensis group, cordate houttuynia group and control group. Spleens were collected for the genetic expression of IFITM3 on 1th day, 3th day, 5th day, and 7th day. Part 2: to study the effects of TCMF with different preparation and method of administration on the genetic expression of IFITM3 in10-day-old 30 broilers in 7 trial days, including TCMF admixture decoction group, TCMF powder group and TCMF admixture after decoction group. Spleens were collected for the genetic expression of IFITM3 on 1th day, 3th day, 5th day, and 7th day. Part 3: to study the effects of TCMF with different doses on the genetic expression of IFITM3 in 7 trial days, including higher TCMF group with 0.5g/kg, medium TCMF group with 1g/kg, lower TCMF group with 2g/kg, toxin blank group and control blank group with each group 30 broilers. To collect spleens for the genetic expression of IFITM3, the contents of IFN, the detoxification amounts and the blood biochemical on 1th day, 3th day, 5th day, and 7th day. The results revealed as follows:1. The genetic expression of IFITM3 in TCMF was higher than others in 3th day, extreme significantly increased with control group(P<0.01) and significantly increased with folium isatidis group(P<0.05) in 5th day. The folium isatidis group was significantly increased with control group(P<0.05) in 5th day. The results showed that the TCMF could improve significantly the genetic expression of IFITM3 with Chinese single medicine.2. The genetic expression of IFITM3 in TCMF admixture decoction group increased with TCMF powder group and significantly increased with TCMF admixture after decoction group(P<0.05) in 5th day. The results showed that the TCMF with admixture decoction could improve significantly the genetic expression of IFITM3 with TCMF admixture after decoction group.3. The TCMF had significantly protective effect in NDV broilers. The protection ratio of higher TCMF group was extreme significantly increased with toxin blank group(P<0.01). With the increase of the concentration of TCMF, the protection ratio of broilers increased.4. The contents of IFN-?, IFN-? and IFN-? in serum in medium TCMF group and lower TCMF group decreased significantly with toxin blank group(P<0.05). The results showed that the TCMF could induce the response of IFN.5. The genetic expression of IFITM3 in TCMF presented negative correlation with the expression of IFN.Conclusion: The TCMF decreased significantly the infection of NDV. With the addition of dosage on TCMF had strong antivirus activity.
