In this study, Congjiang pigs were regard as the research object, and bone morphogenetic protein 15(BMP15) gene and retinoic acid receptor ?(RARG) gene were selected as candidate genes that influenced Congjiang pig reproductive performance. The DNA mixed pool, PCR direct sequencing, PCR-SSCP technique were used to research single nucleotide polymorphisms of RARG and BMP15 gene for Congjiang pigs, looking for genetic markers of having association with reproductive traits. Using real-time quantitative PCR to research RARG and BMP15 gene expression differences on m RNA levels among different organizations for different growth stages Congjiang pigs. The test results were as follows:1.The study on RARG gene showed, 4 SNPs were found among the test groups and constructed 6 kinds of haplotypes. The correlation analysis concluded that C1464 T and C128 T sites had a significant impact on the reproductive performance of Congjiang pigs(P<0.05), and C allele and A allele might be favorable alleles to the total number born, number born alive, teat number of Congjiang pigs.2.The study on BMP15 gene showed, 3 SNPs were found among the test groups and were in linkage disequilibrium, constructed 6 kinds of haplotypes. The correlation analysis concluded that C122 T, A13 G sites of BMP15 gene might be 2 molecular marker which affected reproductive performance of Congjiang pigs, and the haplotype H3, CC genotype, AB genotype, might be favorable haplotype and genotype that influenced number born and teat number of Congjiang pigs.3.Real-time PCR showed, RARG gene and BMP15 gene m RNA were expressed in 15 different tissues for 2, 4, 6, 8, 10 months old Congjiang pig sows, but there were some expression differences between different tissues in different months. Among the ovarie, oviduct, uterine body, uterine horn for Congjiang pig sows, 6 months of ageCongjiang pigs showed higher RARG gene m RNA expression levels than other months of age. But for BMP15 gene, the m RNA relative expression levels of ovarie,oviduct, uterine body, uterine horn for 5 months age stage sow stage all showed higher expression levels than the rest of the organization(P<0.05). And in ovarian tissue, BMP15 gene m RNA expression levels showed gradually increasing trend with the increase months of age of sows.
