Protein content of feather is as high as 90%.Waste feather was produced by the large amount of consumption of poultry.Because of large amounts of disulfide bonds were contained in the feather keratin and worked as a complex 3D mesh structure,Feather has good stability and hard to degrade.Especially,it is hard to be degraded in natural environment.At present,there are physical degradation,chemical degradation,enzyme degradation and microbial degradation to deal with the feather.Although the physical and chemical methods are researched earlier,but they have many shortcomings,such as low degradation efficiency.Use of microbial degradation of the waste feathers become a kind of relatively appropriate choice.There have been many reports show that Bacillus licheniformis which are able to degradate feather are separated from decayed feathers,however,this kinds of Bacillus licheniformis are also need to do a series of safety evaluation before they can be used.In this study,gene of the protease was inseted into pPICZaA,and transformed them into Pichia pastoris.High copy strains was screened.Methanol is used in fermentation.Unfortunately,the supernatant can not degrade feather.We predicted that it maybe the low production of the enzyme and the single enzyme,.On the other hand,high efficiency degradation of feather Bacillus licheniformis are screened from saled probiotics,which can adjust the dysbacteriosis,has the unique biological oxygen-taking mechanism,that can prevent poultry from diseases.Then,optimization of feathers degradation process is designed according to the pretreatment of feather,inoculum size,the composition of culture medium,fermentation time.Original feather is boiled in 0.02%NaOH solution for15 min.10%the ratio of liquid inoculation seeds is added to optimized medium of feathers,culture at 37 ?,pH value is under natural conditions.Amplification experiment was done in 400 L fermentor.Fermentation period is shorten to 70 h since the high dissolved oxygen.B.licheniformis are all in spores state,after disposal of fermented liquid,powder product is prepared,and bacteria content is 4.7×1011CFU/g,protein content is 7.36%.This study created a new use of probiotics with high efficiency of degradation feathers to get new type of feed additives method.
