Rehmannia glutinosa,one of the most famous medicinal plants,is a perennial herbaceous plant of the Scrophulariacea family,the geo-authentic producing areas is “huaiqing prefecture” in Henan,and also has the longest plant history there.Rehmannia glutinosa is one of the famous “four huai medicine”.In the production,the asexual reproduction has lead to the variety degeneration,seriously pests and diseases.So it is necessary to breed new variety with good quality,high yield,high resistance.On the basis of previous study,we use hybridization and space mutation,and has got new strains HuaidiJin 8 and Huaidi 83,one new variety Huaidi 81,and has passed the identification of provincial Agriculture Department.The production,quality and resistance have been contrasted.We established seed breeding and production technical system,the production technologies has been studied,and has significantly economic and social benefit.The main research results as follows.1.The three new strain was get by system selection in the basis of early-hybrid breeding and space breeding,by using the variety comparison test,regional demonstration test and production demonstration test to breeding Huaidi 81 new variety,and has passed the identification of provincial Agriculture Department.Huaidi Jin8 and Huaidi 83 new strains were get by system selection in the basis of early-hybrid breeding,and lay the material foundation for new variety breeding.2.By the experiment of the breeding of new variety.This paper amis to determine component indices of Rehmannia glutinosa Huaidi 81 and five major cultivars,catalpol and acteoside by HPLC,and analyze the difference of the individual plant fresh weight and the contents of index components by SPSS to breeding new cultivar.The results were as follows.The individual plant fresh weight of Huaidi 81 was apparently higher than major cultivars,among them,Huaidi 81>85-5> Jinjiu> Huaifeng> Qinhuai> Beijing No.3.The content of catalpol in Huaidi 81 has no significant difference with Jinjiu,and has high significant difference with other five cultivars,among them,BeijingNo.3(1.601%)>Qinhuai(1.588%)>Huaidi81(1.314%)>Jinjiu(1.277%)>85-5(1.073%)> Huaifeng(0.924%).The content of acteoside in Huaidi 81 has high significant difference with five major cultivars,among them:Huaidi 81(0.096%)> Qinhuai(0.069%)> 85-5(0.047%)> BeijingNo.3(0.035%)> Huaifeng(0.023%)> Jinjiu(0.022%).The conclusion:the comprehensive characters of the individual plant fresh weight and component indices in Huaidi 81 have significant advantages than major cultivars.By the field resistance experiment,we know that Huaidi 81 has high resistance to spot blight high resistance,and has moderate resistance to ring spot.In conclusion,Huaidi 81 has excellent comprehensive characters.3.By the analysis of variety yield,index component,leaf color and photosynthetic characteristics of Rehmannia glutinosa major cultivars,which can provide basis to breed new cultivar.Randomized block design,HPLC,LI-6400 XT photosynthetic are used to messure the production,index component,Photosynthesis characteristic index.The results showed that,the production of HuaidiJin 8 is second in 85-5,that is 70.11kg(10m2),Huaidi 83 is third in 85-5 and HuaidiJin 8,that is 68.09kg(10m2).The content of catalpol and acteoside in HuaidiJin8 are in the third place,that is 1.371%,0.051%,The content of catalpol and acteoside in Huaidi 83 are just higher than Jinjiu,that is 1.212%,0.036%.In the the chlorophyll content of HuaidiJin 8,Huaidi 81,85-5,Jinjiu and Huaidi 83)were higher,the anthocyanin content of Jinjiu,HuaidiJin 8 were higher,Huaidi 83 was the lowest;the photosynthetic rate of Huaidi 81,Huaidi 83 and HuaidiJin 8 were higher;the chlorophyll content was positively correlated with photosynthetic rate,the anthocyanin content was negatively related with photosynthetic rate.The new strain HuaidiJin 8 and Huaidi 83 had good synthetical properties,which were suitable for popularizing in the production properly.4.FTIR combined compare software to analyze eighteen different radix rehmannia.Results:(1)The FTIR fingerprint of radix rehmannia contained thirteen characteristic fingerprint peak and can lay the foundation for the identification of Rehmannia glutinosa from genuine producing area.(2)The similarity of Gansu,Sichuan had big difference with Huai zone,that is their quality is inferior to Huai area.(3)Cluster analysis showed that the radix rehmannia in Huai zone were in the same group,indicating that producing area can distinguish the similarity of Rehmannia glutinosa.No.3 samples in Gansu province,No.4,and No.13 with steam drying were in the same group,that is the processing method had influence to sample quality.The radix rehmannia in Huai zone were in the same group,but Jinjiu,Huaifeng also in the same group,that is varieties have some difference to the quality of Rehmannia glutinosa.Conclusion: The Fingerprints of Radix Rehmannia in Huai zone by FTIR had good stability and reproducibility and were suitable for quality control of Rehmannia glutinosa,and provide the theoretical and technical support for further rationally develop and utilize.5.FTIR combined compare software to analyze ten different radix rehmannia preparata.Results:(1)The FTIR fingerprint of radix rehmannia preparata contained thirteen characteristic fingerprint peak and can provide technical support for deep processing.(2)Different fingerprints had good similarity,that is the major components of radix rehmannia preparata in Huai Zone were similarity.(3)Cluster analysis showed that the same processing method of radix rehmannia preparata were in the same group except No.9 and No.10,indicating that the major components of radix rehmannia preparata in Huai zone were semblable.Maybe No.9 was processed in 2012,then the processing method and time have impact on the quality of radix rehmannia preparata.Conclusion: The study provide a simple,quick and effective means for distinguish different processing method of Rehmannia glutinosa,and provide the theoretical and technical support for deep processing.6.The operating standard of production technologies of Rehmannia glutinosa has been studied,which provide theoretical and technical support for create new varieties and large scale cultivation.
