Newborn piglets diarrhea piglets is a disease with high mortality rates and the important factorswhich can influence the pig production results and economic benefits, the factor that cause diarrhea in addition to the poor management, pathogen infection is an momentous influencing factor.Causing diarrhea piglets pathogen mainly has two elements, namely, virusand bacteria.Viral causes of diarrhea accounted for the vast majority of the newborn piglets diarrhea.Piglet diarrhea disease clinically the original mixed infection phenomenon is relatively common, complicate the clinical symptoms of diarrhea piglets and pathological changes, it is hard to rely on clinical symptoms and pathological changes were diagnosed, which brought great challenges for disease diagnosis and control work.To identify diarrhea piglets related viruses mixed infection situation and the regularity of the onset of diarrhea pigletsin 2014-2015 in shandong province, establish porcine epidemic diarrhea virus laboratory diagnosis quickly and efficiently.Weinvestigate diarrhea piglets farms in shandong province in 2014-2015.In addition, 68 simples of clinical cases of diarrhea wascollected from several cities in shandong province.Using RT-PCR/PCR technique and immunohistochemical technologyproceed related virus detection.In shandong province the results show that theseasonal of the piglet diarrhea is not obvious, the cold winter and spring season multiple transformate for four seasons often hair, and the onset age, had a tendency to expand, the illness also increase significantly.Whichbrings greater challenges for the disease 's prevention and control.Virus detection results suggest that thepositive rate was 88.24%(60/68)during the 68 epidemic materials, percentage of PEDV infection and pathogenesis of alone was 82.35%(56/68), TGEV infection and pathogenesis of separate proportion was 2.94%(2/68), but did not find the Po RV individual cases of infection and disease;Diarrhea virus mixed infection and pathogenesis of 2.94%(2/68).Research results show that the newborn piglets diarrhea in shandong province mainly caused by viral infection, with PEDV infection alone is given priority to, diarrhea virus mixed infection ratio is low.There is low proportion of the TGEV infection and pathogenesis of alone,PEDV has become the leading cause of pig farms in shandong province viral diarrhea piglets.In order to further reveal the PEDV molecular characteristics and molecular epidemiology, providing theoretical basis for prevention and control of PED, outburst of 10 strains PEDV isolates in shandong mainly fiber protein gene(S) total length of the sequence determination and analysis.Results indicate that the length of 10 strains of PEDV S gene is 4161-4170 nt, coding 1387-1390 amino acids.Homology analysis results manifest that the 10 strains of shandong nucleotide homology between isolates(97.9%-99.5%) and amino acid homology(96.9%-98.8%) r;They share nucleotide homology(98.2%-99.4%) and amino acid homology(97.8%-99.1%)with MN form US and HLJ-2012 from CHINA, and the strain of the separation of the domestic early CH/S(1986), Belgium strains CV777 and domestic vaccine strain CV777 nucleotide homology(93.4% 93.9%) and amino acid homology(92.2% 92.9%).Amino acid sequence analysis showed that new isolates and the domestic vaccine strain CV777 compared in more than one site amino acids have a specific replacement and insertion, substitution, and insert the amino acids with the virulence of the virus and the relationship between the antigenic change still needs further research.In shandong province as a whole, significant changes have taken place in the pathogenesis of viral diarrhea piglets rule, PEDV is the foremost reason which cause viral diarrhea.Comparing with other isolates of PEDV S gene has a unique molecular characteristics of the virulence of new isolates In shandong province,and existing vaccines for these strains protective effect remains to be further study.
