Porcine epidemic diarrhea caused by the Porcine epidemic diarrhea virus(PEDV)is one of the typical coronavirus structure.The primary structure of the protein has four,were phosphorylated nucleocapsid protein(N)and membrane protein(M),glycosylation of the spike protein(S)and vesicle membrane protein(E).The S glycoprotein is B lymphocyte antigen determinant of major carriers,is able to induce neutralizing antibodies and provide an important structural protein in immune protective effect,S protein of virus infection,play its pathogenicity and host cell tropism plays a key role.In this paper,the fusion protein expression of S1 PEDV protein at the same time to select nine different epitopes,as well as for the S2 PEDV protein to select five different epitopes.Three ways of antigen using BALB/C mouse monoclonal antibody preparation technology are identified and the corresponding epitope peptide fragment of monoclonal antibody,finally for PEDV S1 expression of fusion protein and PEDV S1 nine different polypeptide epitope bit and PEDV S2 five different peptides table position to obtain the monoclonal antibody effect were analyzed and summarized.Finally,the paper confirmed that the selected peptides of the PEDV S1 and PEDV S2 close to the C-terminal region have good the antigenicity of the polypeptides,monoclonal antibody against the fusion protein was only as antibody titer reference.The research aim is to provide a reference for the development of PEDV vaccine in the future.
