10 wheat varieties(lines)were used as the experimental material,adopted two methods determination of seedling growth and conductivity determination of seed germination index,wheat seeds LOX activity was measured by spectrophotometry,accelerated aging process grain test was used to detect the germination index and storability.Determination of the accelerated aging test material 2-6 days of LOX activity,germination index,activity of some enzymes and conductivity was analyzed.To compare the germination index and storage characteristic differences between wheat varieties,investigate the germination of wheat seed LOX activity and index and storage characteristic relations.The main results were as follows:1,the accelerated aging test results.Using high temperature and high humidity under the condition of accelerated aging,varieties(or lines)and dealing with the analysis results show that the variance between LOX activity and germination rate,MDA,CAT and SOD,soaking liquid conductivity,relative conductivity,root length and fresh weight of nine characters between varieties(or lines)and processing were significant or extremely significant;Absolutely conductivity reached extremely significant difference between varieties(or lines);Buds reached extremely significant difference between the long handle.The 11 characters above,in addition to the absolute conductivity,the rest of the 10 traits of species x treatment reached significant or extremely significant difference were observed in the interactions.LOX activity amplitude without aging treatment(0d)of all varieties(lines)of 17.1-43.5 AU / min ? g,with an average of 26.7 AU / min ? g;Higher LOX activity varieties(lines)Huacheng 3366,lunxuan22,wanmai47 and so on.low as Shannon precocious Department Huacheng 2152,Sumai 188 and so on.In the accelerated aging of 2d,the varieties of LOX activity range 19.87 —38.67 AU/min? g,with an average of 28.4 AU/min? g.Reach 4d in aging,the LOX activity of all varieties(lines)the range of 31.2 —31.2 AU/ min? g,with an average of 38.0 AU/ min? g;And other high LOX activity as Annong 0305,Huacheng 3366 and Luomai 10,lower as Huacheng 2152,Sumai 188 and lunxuan 22 rounds and so on;After accelerated aging 6d,LOX activity range from 17.7 to32.1 AU/ min? g,with an average of 19.5 AU/ min? g.Among them,high LOX activity varieties Annong 0305 and Luomai 10,the lower shannong(later)?shannong(earlier)and Huacheng 2152 and so on.Therefore after the first increased and then decreased LOX activity appears to accelerate the aging process.Untreated(0 d)seed germination rate range from 76.0% to 97.3%,the average was 91.67%,among them 9 varieties(or lines)of untreated seeds germination rate above 85%,the huacheng2152 germination rate was 76.0%.With the accelerated aging treatment process,the germination rate showed a trend of decline.Aging in 2d,the germination rate of range from56.0% to 56.0%,the average is 76.0%;At this point,the higher germination rate for Annong0305 and Huacheng3366 and so on;Lower germination rate of wheat 10 are Shannon(earlier)and so on;Aging 4d,the germination rate of the range of 32.0% to 32.0%,the average of 59.3%.At this point,in addition to Annong 0305,the remaining nine varieties(or lines)of germination rate were reduced to below 70%,Basically lost breeding value,The result show that aging 4 d may be these varieties of seed aging deterioration turning point of time.Aged 6 d,germination rate of the range of 4.67%-55.3%,the average of 14.43%.At this point,a significant deterioration,lost all kinds of breeding value.2,natural aging test results.Natural conditions,varieties(or lines)and dealing with the analysis results show that the variance between bud length,MDA,POD,CAT activity,soaking liquid conductivity difference between the varieties and processing achieves very significant;Germination percentage of the difference between varieties significantly;LOX activity,root length,relative electric conductivity,SOD activity were significant or extremely significant difference between the processing.The above 11 characters,root length,germination rate,POD,CAT,SOD activity,soaking liquid seven properties such as conductivity,relative electric conductivity of species x treatment reached significant or extremely significant difference were observed in the interactions.Initial stage of natural aging treatment(CK),various wheat varieties LOX activity range from17.1 to 43.5 AU/ min? g,the average is 25.01 AU/ min? g;Aging after 3 months,LOX activity range from 9.2 to 39.7 AU/ min? g,the average is 23.12 AU/ min? g;After aging for six months,the LOX activity range from17.5 to 46.5 AU/ min? g,the average is 35.04 AU/ min? g;Aged nine months later,LOX activity range from 22.0 to 61.6 AU/ min? g,the average is 66.83 AU/ min? g;Natural aging after 12 months,LOX activity range from 33.07 to 67.07 AU/ min? g,the average is 45.92 AU/ min? g,Higher LOX activity of varieties are Lunxuan 22 and Luomai10,Low LOX activity of varieties are shannon(earlier),Sumai188 and Huacheng 2152.Initial stage of natural aging treatment(CK),germination rate of wheat varieties range of 76.0% to 76.0%.The average is 89.9%;Natural aging after 3 months,the germination rate of the range of 82.0% to 82.0%,the average is 86.9%;After six months,germination rate of the range of 71.3% to 87.7%,the average is 85.1%;Nine months later,the germination rate of the range of 70.7% to 86.7%,the average is 82.8%;After 12 months,the germination rate of amplitude of 52.7% to 52.7%.Average of 78.3%.At this point,the high germination rate of varieties such as Luomai 10(83.7%),lower germination percentage varieties such as Huacheng 2152(52.7%),etc.3,correlation analysis results show that under the accelerated aging treatment,LOX activity and the germination rate,root length,shoot length and fresh weight,SOD activity was significantly or extremely significantly positive correlation,and has negative correlation with CAT activity?the soaking liquid conductivity?absolute conductivity and relative conductivity.Under the natural aging process,LOX activity related to the germination rate was significantly positively,and soak liquid conductivity,absolutely conductivity were very significant negative correlation and relative electric conductivity.This study combined with the accelerated aging and natural aging process,thought that main LOX activity of seeds germination index and storage of resistance has a positive effect.
