In order to explore the influence of cold stress on immunity performance and the green tea on immune function in chicken.A total of two hundred and eighty 21-d-old Yellow-feathered broilers were randomly divided into control group and experimental treatments.The control group was fed the basal diet,the experimental treatments were fed the diets which contained 0.5%、1% and 2% green tea powder respectively.The immune serum and biochemical indices(IgG、IgA、IgM、LZM、GLB、ALB、STP、MDA、NO、SOD)and mRNA relative expression of IL-2、IL-4 in the spleen、thymus and bursa were measured after exposure of 0、6、12、18、30、78 and 150 h cold stress treatments.The results are as follows:1、34-d-old Yellow-feathered broilers were exposure to cold stress under the temperature(10+/-2 degrees C).The concentrations of chicken serum IgG、IgA、IgM were 17.30±0.68μg/ml、995.77±10.50 ng/ml、682.44 ±17.15ng/ml after 0 h of cold stress.At 6h after cold stress,the concentration of serum IgM was significantly increased to 703.05±9.47ng/ml(P<0.01).At12 h after cold stress,the concentrations of serum IgG and IgA were significantly increased to 21.57±0.84μg/ml and1133.19±38.02ng/ml(P<0.01),while the concentrations were kept at a high level until the end of the trial.Serum LZM、GLB and ALB showed a downward trend after the first rise,and were significantly increased to peaks at 12h、78h after cold stress(the two peaks were : LZM:33.40±1.28U/ml、33.08±1.24 U/ml,GLB:27.45±2.35 mg/ml、26.03±2.34 mg/ml,ALB:1.37±0.14 g/ml、1.29 ±0.14g/ml)(P<0.01).2、The concentration of chicken serum NO was significantly difference between0 h and 12h(P<0.01)after cold stress,the minmum value 19.30±1.50 nmol/ml was occur at 78 h after cold stress;the concentration of MDA showed a fluctuation variation,and were significantly raised to 12.87±1.18 nmol/ml and 12.01±1.46nmol/ml at 6h and 78 h after cold stress;the concentration of serum SOD showed a downward trend after the first rise,the peaks were occur at 30 h and 78 h after cold stress,58.17±4.71 U/ml and 57.30±5.74U/ml respectively(P<0.01).3、The spleen expression level of IL-4 mRNA were significantly raised to2.22±1.09(P<0.05)at 78 h after cold stress,which was 0.38±0.09 at 0h after coldstress,whereas,the spleen expression level of IL-2 mRNA were significantly raised to1.44±0.20 and 1.32±0.30 at 78 h and 150 h after stress;the thymus and bursa expression level of IL-2 and IL-4 mRNA showed a downward trend after the first rise,but the difference was not significant(P>0.05)during the whole period of cold stress.4、The broilers were exposure to cold stress under the temperature(10+/-2degrees C)while adding green tea powder in the diets.The green tea powder had no significantly effect on the chicken serum indexes during the period of cold stress(P>0.05).The spleen expression level of IL-2 mRNA in 0.5% 、 2% adding groups were 1.38±0.53 and 1.21±0.36 respectively at 18 h after cold stress,and were significantly higher than the control group(P<0.01);the bursa expression level of IL-2mRNA in 0.5%、1% adding groups were 1.36±0.50 和 1.32±0.60 respectively and were higher than the control group(P<0.01);the thymus expression level of IL-2mRNA in 0.5%、1% and 2% adding groups were significantly raised to 3.12±0.61、3.14±0.55 and 2.25±0.55 respectively(P<0.01)at 0h after cold stress.The thymus expression level of IL-2 mRNA in 0.5%、1% and 2% adding groups were 3.38±0.62、3.17±0.52 和 2.34±0.42 respectively(P<0.01)at 6h after cold stress,which were significantly higher than the control group(P<0.01);the thymus expression level of IL-4 mRNA in 1% and 2% adding groups were significantly raised to 4.52±1.10 and4.26±1.20 respectively(P<0.01)at 0h after cold stress. |
