| There are lots of chicken bones in China.It is of great significance to enhance the additional value of chicken bones by carrying out research on polysaccharide extraction in chicken bones.In recent years,the role of chondroitin sulfate in the treatment of cardiovascular diseases and arthritis has gained popular acceptance.In order to develop resources for the extraction of chondroitin sulfate and achieve high-value utilization of by-products and improve the degree of deep processing of chicken bones.In this study,we use chicken leg bones as raw materials to analyze its basic nutrient components and the distribution of acidic polysaccharides,and uses alkaline hydrolysis and hot-extraction resin adsorption to extract chondroitin sulfate from chicken leg bones and analyzed the structure of the extractions.Provides a certain theoretical basis and technical support for the use of chicken by-products.The specific results were as follows:1.The result of basic nutrients in the chicken leg bones and Alan blue staining of chicken leg bone,show that acidic polysaccharides mainly exist in the cartilage tissue of the two ends of chicken leg bone,providing the basis for the extraction of chondroitin sulfate from chicken leg bones.The chicken leg bone was divided into two parts,included the ends and middle part of the chicken leg bone,and the chondroitin sulfate was extracted by alkaline with enzymolysis method.The yield of chondroitin sulfate from the two ends of chicken leg bone was 4.25%,and the yield of chondroitin sulfate from middle part of chicken leg bone was 1.32%.After ultrafiltration,the purity can reach 90.71%and 88.59%,respectively.2.The heat-resin adsorption extraction method was investigated to extract chondroitin sulfate from chicken leg bone and the method was optimized by response surface methodology basing on the Box-Behnken design.Optimum conditions of resin dosage(10%),absorption time(4.3 h),eluent concentration(2 M),and eluent time(1.3 h)were obtained and the yield of CS reached to 0.14%.The study found that the effect of various factors on the yield were:resin dosage>adsorption time>eluent concentration>elution time.This method realizes the maximum utilization of chicken bone,which was conducive to the sustainable development of the environment and was suitable for factory production.3.The CS1,CS2,and CS3 were analyzed by ultraviolet spectrum,infrared spectrum,agarose-gel electrophoresis,monosaccharide composition,and disaccharide composition analysis.The UV spectra results showed that the maximum absorption peaks of CS1,CS2,and CS3 all at 210-220 nm;the IR spectrum of chondroitin sulfate samples were basically consistent with the IR spectrum of chondroitin sulfate A standard,and all samples absorbed at 850 cm-1;the chondroitin sulfate samples have the same mobility in agarose gel electrophoresis with chondroitin sulfate A standard;rhamnose and glucuronic acid were the main monosaccharides of chondroitin sulfate;monosulfated disaccharidesΔDi6s andΔDi4s were the main constituent unit of the extracted chondroitin sulfate samples.In summary,the chondroitin sulfate extracted from this experiment were mainly A-type chondroitin sulfate,and combined with a few amino acids form chondroitin sulfate peptide. |
