In order to solve the problems of re-constructed tobacco leaf miscellaneous gas weight,lack of flavor and poor taste,this topic uses a combination of heat-resistant enzymes in the tobacco extraction process,in the process degraded proteins,starch and pectin molecules,promoted reconstitution The physical properties such as color of tobacco leaves and tensile strength were increased.At the same time,the contents of aroma substances such as furfural and sterols were increased,which achieved the purpose of upgrading the quality and flavor of reconstituted tobacco.For this purpose,the following researches were mainly carried out: The heat-resistant enzyme was screened through Genbank.The gene fragments of the nature are sequence-optimized,cloned and expressed and purified to collect heat-resistant enzymes;the heat-resistant enzyme is applied to the lixiviation phase of the reconstituted tobacco leaf production process according to the seven schemes of single enzyme,dual enzyme,and three enzymes respectively.With reference to the blank control without heat-resistant enzyme added,the changes of protein,starch,pectin and other macromolecules content in the extract during the extraction process were detected at different time intervals by conventional methods,and detected by GC/MS.Aroma components of extracts and their control;Determination of aroma components by reconstituted tobacco leaves after preparation of finished product and physical indicators for the detection of reconstituted tobacco The reconstituted tobacco leaves prepared on the basis of the blend of finished cigarettes plus 5% of the added volume for sensory quality evaluation.The results showed that:(1)Three kinds of heat-resistant enzymes that could be used in the reprocessing process of tobacco leaves were screened by enzymological properties,including heat-resistant protease(ZZQGY-E01),heat-resistant amylase(ZZQGY-E02),and thermostable pectinase.(ZZQGY-E03)for application.(2)The chemical index and aroma substance content in the extraction of tobacco leaf raw materials were assisted by the 7-item scheme,and it was concluded that the synergistic effect of the three enzymes was the best,leading to a 32% protein degradation,36.4% starch reduction,and 29% reduction in pectin.The amino acid in the extract increased by 7.15 times and the reducing sugar increased by 28%.(3)The use of heat-resistant protease,amylase,and pectinase tri-enzymes to assist the reconstituted tobacco leaves at the extraction stage to prepare reconstituted tobacco products.Some aroma components were found to increase,for example,ionone increased by 17.5% and furfural increased by 41.2.And sensory evaluation results by blending cigarettes showed that sensory-assisted reconstituted tobacco had a 1.0-point increase in sensory evaluation.Based on the above research,we have created a composite heat-resistant protease,amylase,and pectinase to work together in the tobacco extraction process.The tensile strength and the burning rate of the reconstituted tobacco are all increased by 10%.Some of the aroma substances such as sterols,2-The content of acetyl furan,etc.increases,therefore,the process technology achieves the purpose of upgrading the quality and flavor,and is expected to increase its proportion in the cigarette formulation,which has significant economic benefits.
