Sesame contains a variety of nutrients,with high nutritional value and edible functional value,and its use is more extensive.However,white sesame,which is also pedaliaceae,has no record of medical value in many medical collections.Moreover,people always think that black sesame is more beneficial to human health than other sesame seeds such as white sesame.This is only a traditional understanding,or can find the basis of science? No one has systematically research on these issues.Therefore,44 white sesame seeds and 40 black sesame seeds were used as samples to investigate the components and functional components in black and white sesame seeds,determine the antioxidant activity of black and white sesame extract,study the anti-HepG2 cell proliferation of black and white sesame extract,and delay the aging of 2BS cells.Animal experiments were used to evaluate the hypolipidemic effect of black and white sesame on SD rats.(1)With 44 white sesame seeds and 40 black sesame seeds as the research object,the composition of black and white sesame seeds was analyzed.The results showed that there were significant differences in the apparent structure of black sesame and white sesame.The content of dermis,ash,crude fiber,oxalic acid,linoleic acid,and lysine in black sesame was higher than white sesame seeds at the rate of 38.4,23.9,27.4,184.0,4.51,and 7.59,respectively.The content of crude fat,sesamin,and oleic acid in white sesame was higher than black sesame seeds at the rate of 4.9,31.9and 4.9,respectively.In addition to Na,the average content of minerals of black sesame seeds was higher than that of white sesame seeds.The average content of Fe and Ca in black sesame was 2.12 and 1.55 times that of white sesame.(2)Total antioxidant capacity,DPPH radical scavenging rate,and ·OH scavenging rate were used as evaluation indicators for in vitro antioxidant activityThe results showed that the total phenolic content of black sesame was higher than that of white sesame,and the total phenolic content in seed coat of sesame was highest.The content of sesamin in white sesame seeds and the kernel of white sesame seeds was higher than that of black sesame seeds and the kernel of black sesame seeds.The total antioxidant capacity and DPPH radical and·OH clearance of black sesame was higher than those of white sesame 63.2%,46% and 15.2%,respectively.The content of total phenols in black and white sesame and seed coat of black and white sesame was significantly positively correlated with the total antioxidant capacity and DPPH free radical clearance(P<0.01),but the content of sesamin in black and white sesame seeds was significantly negatively correlated with total antioxidant capacity,DPPH and OH free radical clearance.The content of sesamin content in the kernel of white and black sesame seeds was positively correlated with total antioxidant capacity(P<0.05).There was a significant positive correlation between sesamin and sesamolin in black and white sesame and seed coat of black and white sesame(P<0.05).(3)The effects of black and white sesame seeds and the kernel of black and white sesame seeds extract on the proliferation of HepG2 cells and delaying the aging of 2BS cells were studiedThe results showed that the effects of black and white sesame and kernel of black and white sesame extract on the proliferation of HepG2 cells were dose-dependent,and the anti-proliferative ability of black and white sesame was stronger;the effects of black and white sesame and kernel of black and white sesame extract on the T-AOC of 2BS cells all increased with the increase of the extract concentration,and the influence of the black and white sesame extract was greater than that of the kernel of black and white sesame extract.The effect of the black sesame and the kernel of black sesame extract was greater than that of white sesame and the kernel of white sesame;the effect of black and white sesame extract on the MDA content of 2BS cells was first decreased,then increased,then decreased with the increase of the concentration.(4)The model of hyperlipidemic SD rats was used to evaluate the hypolipidemic and antioxidative effects of black and white sesame and the kernel of black and white sesameThe results showed that SD rats fed with high-fat diet for 9 weeks had the same final body weight of white sesame and the kernel of white sesame than black sesame and the kernel of black sesame.The body weight was lower than that of the kernel of black and white sesame group;the liver index was significantly(P<0.05)lower in the normal control group and the experimental group than in the model control group;black and white sesame seeds and the kernel of black and white sesame were able to reduce the content of TC,TG of the serum and reduce AST,ALP enzyme activity;normal control group and each experimental group rats liver MDA were significantly(P <0.05)lower than the model control group;model control group rat liver T-AOC,HDL,GSH-Px,T-SOD was significantly(P<0.05)lower than that of the normal control group,and the T-AOC,HDL,GSH-Px,and T-SOD in the liver of each experimental group were significantly higher than those in the model control group;combined with pathological section analysis,we can see that In addition,black and white sesame seeds and the kernel of black and white sesame can improve the liver's fatty lesions and lipid metabolism disorders and reduce liver lipid deposition,which can protect the liver from damage,so as to reduce the risk of fatty liver.
