The Research On Immune Activity Of Birch Polysaccharides And Beverage Production

Inonotus obliquus causes widespread attention from scholars all over the world because of their functional activity,which is considered as a wonder drug in Russian folk.This paper mainly studies the efficient extraction method of polysaccharides in birch,the separation and purification of polysaccharides in birch,identification of immune activity and production technology of polysaccharides in birch beverage.Specific research contents and results are as follows:The effect of microwave assisted on the extraction rate of polysaccharide was studied.The fungus of the birch is crushed before polysaccharides extraction so that the contact area of raw material and solvent is increased.We can explore the influence of mechanical pulverization and microwave assisted on the extraction rate of polysaccharides through it.We choose three important factors in the extraction of polysaccharide,use the response surface for process optimization to get the best process parameters for: microwave power 467 W,liquid-to-solid ratio20:1,the extraction time 196 minutes.Explore separation and purification of polysaccharides.Separation mainly refers to degreasing,deproteinization,decolorizing and dialysis.We used four kinds of deproteinization methods and made comparison,the maximum of the method was Sevage 86.92%,the method TCA(Trichloroacetic acid)was 53.07%,the enzyme method was 85.63%,and the Sevage+enzyme method was 89.27%.Then we purified polysaccharides by DEAE cellulose column chromatography and gel chromatography,IOP-Ⅰ、IOP-Ⅱ、IOP-Ⅲ、IOP-ⅠA、IOP-ⅠB was obtained.Identify immune activity of polysaccharide IOP-ⅠA.It studies the effects of polysaccharide on the immune organs,the effect on nonspecific immune cells by organ index test,clearance rate of charcoal particles index and macrophage phagocytosis test,so as to identify the immune activity of polysaccharide.The experimental results show that the polysaccharides dose group could significantly increase the cyclophosphamide treatment of viscera index in mice,macrophage phagocytosis index also increases,which shows that polysaccharides have immunological activity.According to the experimental results that birch polysaccharides have immune activity,we will study the production technology of birch beverage which makes polysaccharides to be the functional so as to identify the optimum production technology of birch beverage.The experiment shows that :the best production technology recipe is that: 0.40% birch polysaccharides,0.10% citric acid,10% soft sugar,0.40% CMC.
