Semen Zizyphi Spinosae is Rhamnaceae plants wild jujube's dry mature seeds.It is a kind of traditional Chinese medicinal materials,the main chemical composition can be divided into four categories:triterpene and saponins,alkaloids,flavonoids and polysaccharides,commonly used in virtual vexed sleepless,horrified to the treatment of diseases such as have many dreams,top grade by people as a meditation,sedative.There are a variety of nutrients and active ingredients in Semen Zizyphi Spinosae,therefore,of Semen Zizyphi Spinosae resources development and utilization have the very high economic value.The research results show that the various components of Semen Zizyphi Spinosae have significant effect on reducing the spontaneous activity in mice and have some central inhibitory effect.So the purpose of this study is to fully as much as possible to extract the effective components from Semen Zizyphi Spinosae.This reseatch main studied the optimum extraction conditions of saponins and polysaccharide in Semen Zizyphi Spinosae,after this we take the validation on saponins and polysaccharide's test method.Specific work is as follows:1.Studied the process conditions which used alcohol refluxing method to extract total saponins from Semen Zizyphi Spinosae.The orthogonal test methodology was applied to optimized conditions further. The result showed that the best extraction process as follows:85% ethanol reflux extracting 2 times,ratio of material to liquid 8: 1and 1.5 hours each time.2.An orthogonal test was performed to investigate the influences of the amount of water,decocting time,and times of decoction on the content of polysaccharide.The result showed that the best extraction process as follows: adding water (7 times),and extracting 2 times (1.5h each time).3.The RSD of precision experimental result to test saponins by ultraviolet spectrophotometry is 0.62%,the RSD of reproducibility test result is 0.48%.In recovery rate experimental result,the average recover rate R is 95.75%,the RSD is 1.93%.These results indicate the precision and reproducibility in this method is fine.4.The RSD of precision experimental results to test polysaccharide by ultraviolet spectrophotometry is 0.50%,the RSD of reproducibility test result is 0.58%.In recovery rate experimental result,the average recover rate R is 97.91%,the RSD is 1.75%.These results indicate the precision and reproducibility in this method is fine.
